Thesis The research described in this thesis was performed at Leiden University Medical Center, Department of Medical Microbiology, Leiden, The Netherlands

Financial support Work in the group of WKS was supported by a Vidi Fellowship (864.10.003) of the Netherlands Organization for Scientific Research (NWO) and a Gisela Thier Fellowship from the Leiden University Medical Center.

Cover António Paiva design. Microscopy picture of C. difficile strain RD16, harbouring HupA-HaloTag, induced at mid-exponential growth phase with 200 ng/ml anhydrotetracycline for 1 hour, imaged with the A4 filter cube (340-380 nm excitation and 450-490 nm emission) for the coumarin substrate (at 150 nM) visualization, on a Leica AF6000 microscope with LAS AF software. Microscopy performed by Roxanne Douwes.
