PROPOSITIONS

Belonging to the PhD thesis

The Roles of Noncoding RNAs in B-cell Lymphomas

1. High-throughput based gain- and loss-of-function screens are effective tools to identify B-cell lymphoma B-cell growth-related non-coding RNAs (this thesis).

2. MiRNA expression patterns in Burkitt lymphoma and germinal center B-cells are quite similar, especially for the highly abundant miRNAs (this thesis). 3. MiR-378a-3p is a highly abundant miRNA with increased expression in Burkitt lymphoma

which promotes cell growth by regulating targets such as IRAK4, FOXP1, MNT and JPX (this thesis).

4. MiR-26b-5p suppresses Burkitt lymphoma cell growth via targeting, amongst others, KPNA2 which leads to decreased MYC expression (this thesis and Jie L et al. Cancer Research, 2018).

5. Combining Ago2 RNA-Immunoprecipitation (RIP) with a genome wide CRISPR/Cas9 dropout screen is a powerful approach to identify miRNA targets with effects on B-cell lymphoma cell growth (this thesis).

6. MAFG-AS1, a MYC-induced lncRNA with a predominant nuclear localization, promotes Burkitt lymphoma cell growth (this thesis).

7. Non-coding RNAs show great potential as prognostic markers, biomarkers, or therapeutic targets (Eleni A et al. Nature, 2018).

8. Being prepared before raining and digging a well before thirsty (Zhuzi's Family Motto). 宜未雨而绸缪,毋临渴而掘井。-《朱子家训》

9. Don't forget why we started because we went too far (Kahlil Gibran). 不要因为走得太远,而忘了我们为什么出发。-纪伯伦

10. Where there is a will, there is a way; what you have strived, will be finally repaid (Pu Songling). 有志者事竟成,破釜沉舟,百二秦关终属楚;苦心人天不负,卧薪尝胆,三千越甲可吞 吴。 -蒲松龄

A more comprehensive study, using serial analysis of gene expres- sion (SAGE) and micro-array approaches to compare cHL cell lines with several normal B cell populations, revealed

All cHL cell lines show an increased CN in this region (indicated by BACs above the 0.4 cut-off dashed line). The c-REL gene, mapping within this SRO does not show an increase

To identify miRNAs relevant for BL cell growth, we conducted a screen with miRNA overexpression and inhibition libraries in the ST486 and DG75 cells (Figure 1A).. The

In Chapter 2, we identified miR-21-5p as a miRNA that supports growth of cHL cells. To explore the functional mechanism, we identified potential miR-21-5p target genes based on

In hoofdstuk 4 hebben we een miRNA overexpressie (pool van 44 constructen) en inhibitie (58 constructen) screen gedaan in BL-cellijnen. De effecten op de groei van

Most of the miRNAs highly abundant in germinal center B cells are also highly abundant in Hodgkin lymphoma (This thesis). High-throughput microRNA inhibition screenings

Comparison of the lncRNAs and mRNAs differentially expressed between the normal B-cell subsets and between cHL cell lines and GC-B cells revealed a limited overlap of
