Acknowledgements

I realize that standing in this line of a PhD track is not possible without people around for support. Therefore I would like to express my sincere and heartfelt gratitude to everyone who helped me in any way to reach this goal.

First of all, I would like to thank my promoters (Prof. Dirk J. Slotboom and Dr. Albert Guskov) who fully supported me to build my scientific way, without whom I would not be where I am today. Dirk, you are the most contributed one in my PhD life. I understand how you treat me as your PhD student, to let me grow up. I like the way you supervise me with open minded, constructive critics, ambitious, but friendly and wise approaches. As an Indonesian, I was not used to a direct culture as in the North of the Netherlands, however it is the best way to drive me to reach the finish line.  At the end of my PhD, I had a hard time performing well due to pregnancy and giving birth, however, you still support me without demanding the result. I am so sorry, I did not manage to get a new conformation of the ECF transporters. There is also not enough time to finish the rest of the project and learn many things from you, if there is still an opportunity, I would like to do it more. In other words, I am very grateful to have you as my supervisor. Once again, Bedankt voor alles!

Albert, I am also very grateful that you gave me a chance to learn how to do crystallization and for adopting me as an associated member of your group. I have learned a lot how to set up crystals and solve the structure, I loved to do it and wanted to have more diffracted crystals. Спасибо за поддержку!

Also for Prof. Bert Poolman, thank you for your questions and suggestions during group meetings and for your interest! Your knowledge on science impresses me. For the committee members, Prof. A.S.S. Dömling, Prof. G. Maglia and Prof. D. Fotiadis thank you for your time and energy that you spent on reading my PhD thesis.

My paranimfs, Zaid and Aike. Thank you for your support and kind help. Zaid, you are highly motivated and persistent, and did a lot of experiments when you  were working on the project of ECF transporters, I am sure your PhD will be brighter than mine. For Aike, I enjoyed discussing everything with you about everything, you are humble and friendly. Thank you for translating my summary in Dutch!

I cannot omit the invaluable help and long conversations from Gea, Inneke, Magriet, Renate and Sandra. Especially thank you to Gea, for introducing me to Settlers group. I super enjoyed our board-game night and the food!. Also to Masha and Li Yue, thank you very much for the kind support during my PhD times. Masha, I enjoyed talking and walking along with you and to bring Emery outside. Li Yue, I am so sorry, I could not manage to characterize all the nanobodies, hopefully you will find an interesting one.

I want to thank all past and present Membrane Enzymology and Crystallography members for many chats in the coffee corner, suggestions during group meetings, interactions, activities and the great and pleasant working atmosphere that all of you created in the last 4.5 years. I could not state one by one here because everyone is special to me. Let me know if you visit Indonesia one day.     Also for my collaborators: Weronika, Eleonora, Spyrus and Marysia. Thank you so much for your hardwork!! For Lotteke who spent your valuable time to teach me in the lab when I started my PhD, also for them showing me to solve crystals, Tjaard and Nicola, thank you very much for your kind help and time. I am so sorry Tjaard, I could not manage to say goodbye to you before my departure in Groningen. I will always remember your advice, tips and tricks to solve the structure by hand using ‘’Coot’’.

I also received the emotional support from Indonesian community in Groningen (My family in Kajuit, Planetenlaan, the Noordeplantsoen and around).

Special support came also from my colleagues in the Departments of Biochemistry, IPB in Bogor, Indonesia. Terima kasih atas dukungan dari Bapak dan Ibu dosen and tendik yang memberikan saya keluangan waktu plus-plus untuk menyelesaikan thesis PhD ini.

Finally to my family, without you I would not have been where I am now. You have given me so much inspiration, strength and belief on the way. To my beloved husband, Feri Rahmadi, I am truly grateful for the countless number of small and big adventures that we enjoyed together. I admire you and am inspired by your drive and persistence. Even though taking care of our baby in Groningen has not always been easy, we made it through, and I believe that it has only made us stronger!

To my little boy, Emery, you are my angel. I am very grateful to have you during my PhD times. The smile, laugh, mumbling and every single action from you cheered me up when I was so stressed to finish my thesis. Absolutely, you energize me to finish writing. I hope you will read my thesis in the future and inspire you to dive into the world of science. Also for Abang Azam, Bang Askari, Bang Al terspesial, Aa Aflah dan teteh Katsuni, semoga thesis ini menginspirasi kalian menggapai cita-cita kelak. Hopefully you will be the next persistent and patient generations!!!
