Total synthesis of natural products has over many decades driven the progress in the field of organic synthesis and allowed to push the limits of synthetic methodology.
There are various incentives to embark on the total synthesis of a natural product:
×
Synthesis of the proposed structure of a natural product is the golden standard for the confirmation of its chemical structure and (absolute) stereochemistry. It allows direct comparison of natural and synthetic material by spectroscopic and chromatographic methods.×
Asymmetric synthesis of complex natural products serves as excellent training for an organic chemist, from retrosynthetic analysis to reaction optimization.×
Total synthesis is also of great value to the field of organic synthesis as it aids in identifying the limits of the current chemical methodology.×
Total synthesis has over the years served as a driving force for the development of new chemical transformations, often in asymmetric fashion, to access complex structural motifs.×
Total synthesis of natural products provides synthetic reference material for the assessment of their biological activity. As synthetic reference material is free of biologically active contaminants originating from isolation, it enables biological studies with minimal risk of false positive or false negative results.In chapters 2, 3 and 4, the asymmetric total synthesis of the proposed structure of the tricyclic biphytanediol of the archaeal membrane lipid
crenarchaeol is described. This target molecule contains a complex 5-6-ring system, which, to this date, was never accessed synthetically. In order to achieve the synthesis of the target biphytanediol, new catalytic asymmetric methodology was developed (chapter 3). This illustrates the importance of total synthesis to identify and push the limits of current synthetic methods. The asymmetric synthesis of the target biphytanediol enabled the comparison with natural crenarchaeol. This was done by means of chemical derivatization of natural and synthetic material followed by GC-MS analysis. In addition, the synthetic biphytanediol was directly compared to natural crenarchaeol by NMR analysis (chapter 4). Thorough analysis revealed a mismatch in chemical structure of the synthetic material and natural crenarchaeol and ultimately allowed the proposal of a revised structure of crenarchaeol beyond reasonable doubt. This finding highlights the necessity of total synthesis for the definite confirmation of the chemical structure and stereochemistry of natural products.
Chapters 5 and 6 describe the first asymmetric total synthesis of the mycobacterial cell well wall diacyl trehaloses DAT1, DAT2 and DAT3. The
immunostimulatory properties of isolated natural DAT have been studied, yet as a mixture rather than as single discrete molecular species. After successful total synthesis of these three trehalose glycolipids DAT1, DAT2 and DAT3,
they were studied for their potential to activate the macrophage inducible Ca2+-dependent lectin receptor (Mincle) which is part of the innate immune
system. Significant differences of Mincle binding and activation were found, depending on the nature of the chiral fatty acid ester moieties. HPLC-MS analysis after co-injection of the synthetic DATs and cell wall extracts of
Mycobacterium tuberculosis revealed that the proposed structure of DAT2 (as
in the synthetic material) does not correspond to natural DAT2. This highlights
the value of total synthesis for the assessment of bioactivity of natural products and – again – for the confirmation of chemical structure.
Building on the total synthesis of DAT, chapter 7 describes the first asymmetric total synthesis of the mycobacterial cell wall glycolipid pentaacyl trehalose (PAT). This significantly more complex ‘big brother’ of DAT bears a highly non-symmetrical acylation pattern. Thus, a suitably-protected trehalose intermediate was prepared by glycosylation rather than desymmetrization of trehalose itself. The stereoselective construction of the challenging a,a-configured 1,1’-glycosidic linkage was achieved by careful optimization of the glycosylation conditions. The overall step count of the total
synthesis was kept to a minimum by the installation of four out of five ester moieties in one step. After having achieved synthetic access to PAT, comparison to the NMR spectra of the natural material unveiled a mismatch between the structure of synthetic PAT and isolated natural PAT, which is attributed to a different acylation pattern. Therefore, the proposed structure of natural PAT has to be revised. This again exemplifies the importance of total synthesis for the confirmation, or revision, of the chemical structure of natural products. Further research is ongoing to unravel the exact chemical structure of naturally occurring PAT.

De afgelopen decennia heeft de totaalsynthese van natuurproducten voor vooruitgang gezorgd op het gebied van organische synthese en daarmee ook de grenzen van de synthetische methodologie verlengd.
Er zijn verschillende redenen om aan de totaalsynthese van een natuurproduct te beginnen:

De synthese van een voorgestelde structuur van een natuurproduct is de gouden standaard voor de bevestiging van de chemische structuur en de (absolute) stereochemie. Hierdoor is het mogelijk om een directe vergelijking te maken tussen het natuurlijke en het synthetische materiaal met behulp van spectroscopie en chromatografie.×
De asymmetrische synthese van complexe natuurproducten is een uitstekende training voor een organisch chemicus, van retrosynthetische analyse tot de optimalisatie van een reactie.×
De totaalsynthese is van grote waarde voor de organische chemie doordat de limitaties van de huidige chemische methodologie duidelijker worden.×
Door de jaren heen heeft de totaalsynthese als een drijvende kracht gediend voor de ontwikkeling van nieuwe chemische transformaties, vaak op asymmetrische wijze, om complexe structurele motieven te verkrijgen.× De totaalsynthese van natuurproducten zorgt voor synthetisch mteriaal dat als referentie gebruikt kan worden bij het vaststellen van de biologische activiteit. Omdat synthetisch referentiemateriaal geen
biologisch actieve verontreinigingen bevat, wordt het risico op foutpositieve of foutnegatieve resultaten van een biologische studie verkleind.
In hoofdstuk 2, 3 en 4 wordt de totaalsynthese van de voorgestelde structuur van het tricyclische bifytaandiol van het archaïsche membraan lipide crenarchaeol beschreven. Dit doelmolecuul bevat een complex 5-6-ringsysteem, dat tot nu toe nog nooit is gesynthetiseerd. Nieuwe katalytische asymmetrische methodes zijn ontworpen om het bifytaandiol te maken (hoofdstuk 3). Dit toont aan hoe belangrijk de totaalsynthese is om de grenzen van de huidige synthetische methoden te identificeren en te verleggen. Door de asymmetrische synthese kon het verkregen doelmolecuul bifytaandiol vergeleken worden met het natuurlijk verkregen crenarchaeol. Dit werd gedaan door middel van chemische derivatisering van natuurlijk en synthetisch materiaal gevolgd door GC-MS analyse. Bovendien werd het synthetische bifytaandiol rechtstreeks vergeleken met het natuurlijke crenarchaeol met behulp van NMR analyse (hoofdstuk 4). Grondige analyse bracht een mismatch aan het licht tussen de chemische structuur van het synthetisch materiaal en het natuurlijk verkregen crenarchaeol waardoor het voorstel van een herziende structuur van crenarchaeol zonder enige twijfel mogelijk gemaakt kon worden. Deze bevinding benadrukt de noodzaak van totaalsynthese voor de definitieve bevestiging van de chemische structuur en stereochemie van natuurproducten.
Hoofdstukken 5 en 6 beschrijven de eerste asymmetrische totaalsynthese van het mycobacteriële celwand diacyl trehalosen DAT1, DAT2 en DAT3. De
immunostimulerende eigenschappen van geisoleerd natuurlijk verkregen DAT zijn bestudeerd als mengsel van moleculen in plaats van elk molecuul afzonderlijk. Na de succesvolle totaalsynthese van de drie trehalose glycolipides DAT1, DAT2 en DAT3, werd hun potentie om de door macrofagen
induceerbare Ca2+-afhankelijke lectine receptor (Mincle), dat een onderdeel
is van het aspecifieke immuunsysteem, te activeren. Hierbij werden significante verschillen in binding aan Mincle en activering gevonden die afhankelijk zijn van de aard van de chirale vetzuurestergroepen. HPLC-MS analyse na co-injectie van de synthetische DATs en de celwand-extracten van Mycobacterium tuberculosis toonde aan dat de voorgestelde structuur van DAT2 (zoals in het syntetisch materiaal) niet overeenkomt met het
beoordeling van de biologische activiteit van natuurproducten en – nogmaals – voor de bevestiging van de chemische structuur.
Voortbouwend op de totale synthese van DAT, wordt in hoofdstuk 7 de eerste asymmetrische totaalsynthese van de mycobacteriële celwand glycolipide pentaacyl trehalose (PAT) beschreven. Deze aanzienlijk complexere ‘grote broer’ van DAT heeft een sterk asymmetrisch acyleringspatroon. Zodoende werd een geschikt beschermd trehalose-tussenproduct gesynthetiseerd door glycosylering in plaats van desymmetrisering van trehalose zelf. De stereoselectieve constructie van de uitdagende a,a--geconfigureerde 1,1’-glycosidebinding werd bereikt door zorgvuldige optimalisatie van de glycosyleringscondities. Het totale aantal stappen van de totaalsynthese werd tot een minimum beperkt door de installatie van vier van de vijf estergroepen in één stap.
Na het verkrijgen van synthetisch PAT, onthulde vergelijking met de NMR-spectra van het natuurlijk materiaal een mismatch tussen de structuur van synthetisch verkregen PAT en geïsoleerd natuurlijk PAT, dat wordt toegeschreven aan een ander acyleringspatroon. Daarom moet de voorgestelde structuur van natuurlijk PAT worden herzien. Dit illustreert opnieuw het belang van totaalsynthese voor de bevestiging of herziening van de chemische structuur van natuurproducten. Verder onderzoek is gaande om de exacte chemische structuur van natuurlijk voorkomende PAT te ontrafelen.
Deutsche Zusammenfassung
Die Totalsynthese von Naturstoffen hat über viele Jahrzehnte den Fortschritt auf dem Gebiet der organischen Synthese vorangetrieben und es ermöglicht, die Grenzen der Synthesemethodik zu verschieben.
Es gibt unterschiedlichste Gründe, die Totalsynthese eines Naturstoffs in Angriff zu nehmen:
× Die Synthese der vorgeschlagenen Struktur eines Naturstoffs ist der Goldstandard für die Bestätigung oder Revidierung der chemischen Struktur sowie der (absoluten) Stereochemie. Sie ermöglicht den direkten Vergleich von natürlichem und synthetischem Material mit Hilfe spektroskopischer und chromatographischer Methoden. × Die stereoselektive Synthese komplexer Naturstoffe dient nicht nur
als hervorragende Ausbildung für jeden organischen Chemiker, sondern identifiziert auch die Grenzen der aktuellen chemischen Methodik.
× Die Totalsynthese hat im Laufe der Jahre als treibende Kraft für die Entwicklung neuer chemischer Transformationen – oftmals stereoselektiv – für Herstellung komplexer Strukturmotive gedient. × Die Totalsynthese von Naturstoffen liefert synthetisches Material zur
Studie von deren biologischer Aktivität. Dieses Referenzmaterial ermöglicht biologische Studien mit minimalem Risiko für falsch positive oder negative Ergebnisse, da es frei von anderen biologisch aktiven Verunreinigungen ist, die aus der Isolierung stammen. In den Kapiteln 2, 3 und 4 wird die asymmetrische Totalsynthese der vorgeschlagenen Struktur des trizyklischen Biphytandiols von Crenarchaeol beschrieben, ein Membranbestandteil mariner Archaeen. Dieses Zielmolekül
enthält ein komplexes 5-6-Ringsystem, das bis heute nie synthetisch hergestellt wurde. Um die Synthese des Bihpytandiols erfolgreich durchzuführen, wurde eine neue katalytische and stereoselektive Methode entwickelt (Kapitel 3). Dies zeigt, wie wichtig die Totalsynthese ist, um die Grenzen der derzeitigen Synthesemethoden zu identifizieren und zu überschreiten. Die asymmetrische Synthese dieses Biphytandiols ermöglichte den Vergleich mit natürlichem Crenarchaeol mittels Derivatisierung und GC-MS-Analyse sowie direktem NMR-Vergleich (Kapitel 4). Eine detaillierte Analyse ergab die Nichtübereinstimmung der chemischen Struktur des synthetischen Materials und natürlichem Crenarchaeol. Dies ermöglichte letztendlich den Vorschlag einer überarbeiteten Struktur von Crenarchaeol. Diese Erkenntnis unterstreicht die Notwendigkeit der Totalsynthese zur eindeutigen Bestätigung der chemischen Struktur und Stereochemie von Naturstoffen.
Kapitel 6 beschreibt die erste stereoselektive Totalsynthese der Diacyl Trehalosen DAT1, DAT2 und DAT3, welche Bestandteile der Zellwand von
Mycobacterium tuberculosis sind. Die immunstimulatorischen Eigenschaften
von isoliertem natürlichem DAT wurden untersucht, jedoch als Mischung verschiedener DATs und nicht als separate, molekulare Struk wurden sie auf ihr Potenzial untersucht, den durch Makrophagen induzierbaren Ca2+
-abhängigen Lektinrezeptor (Mincle) als Teil des angeborenen Immunsystems zu aktivieren. Abhängig von der molekularen Struktur der chiralen Fettsäureestereinheiten wurden signifikante Unterschiede der Mincle-Bindung und -Aktivierung gefunden. Die HPLC-MS-Analyse nach Co-Injektion der synthetischen DATs und Zellwandextrakte von Mycobacterium
tuberculosis ergab jedoch, dass die vorgeschlagene Struktur von DAT2 (wie
im synthetischen Material) nicht dem von natürlichem DAT2 entspricht. Dies
unterstreicht den Wert der Totalsynthese für die Bewertung der Bioaktivität von Naturstoffen und - wiederum - für die Bestätigung der chemischen Struktur.
Aufbauend auf der Totalsynthese von DAT beschreibt Kapitel 7 die erste stereoselektive Totalsynthese des mykobakteriellen Zellwandglykolipids Pentaacyl Trehalose (PAT). Dieser wesentlich komplexere „große Bruder“ von DAT weist ein stark unsymmetrisches Esterifizierungsmuster auf. Somit wurde ein geeignet geschütztes Trehalose-Zwischenprodukt eher durch Glykosylierung als durch Desymmetrisierung von Trehalose selbst hergestellt. Die stereoselektive Darstellung der synthetisch anspruchsvollen
a,a-konfigurierten 1,1'-glycosidischen Bindung wurde durch sorgfältige Optimierung der Glycosylierungsbedingungen erreicht. Die Gesamtschrittzahl der Totalsynthese wurde durch die simultane Synthese von vier von fünf Estereinheiten in einem einzigen Schritt minimal gehalten. Nach erfolgreicher Synthese von PAT ergab der Vergleich mit den NMR-Spektren des natürlichen Materials eine Abweichung in der Struktur von synthetischem PAT und isoliertem natürlichem PAT. Dieser Unterschied zum ursprünglichen Strukturvorschlag wird einem anderen Acylierungsmuster von natürlichem PAT zugeschrieben. Dies zeigt erneut die Bedeutung der Totalsynthese für die Bestätigung oder Revidierung der chemischen Struktur von Naturstoffen, was letztendlich die Korrektur der aktuellen Literatur ermöglicht. Weitere Forschung ist im Gange, um die genaue chemische Struktur von natürlich vorkommendem PAT zu entschlüsseln.
