V O O R W O O R D
P r o e f s c h r i f t e n worden in het algemeen nauwelijks gelezen. In de meeste gevallen beperkt men zich tot de laatste s t e l l i n g , het dankwoord en de l e v e n s l o o p . Hiervoor zijn 3 (goede) redenen aan te wi jzen:
1) de tekst is meestal in het Engels g e s t e l d , 2) het onderwerp is erg gespecialiseerd en zeker
voor een leek nauwelijks te b e g r i j p e n , 3) w e t e n s c h a p p e l i j k e teksten zijn saai.
In dit gedeelte van het p r o e f s c h r i f t wilde ik vertellen wat ik gedurende 3 jaar met "die k a r p e r s " gedaan heb. De tekst is in het n e d e r l a n d s , verder heb ik geprobeerd ook het 2e en 3e argument te o n d e r v a n g e n .
Ik hoop dat U na het lezen van dit gedeelte met mij de volgende stelling kunt o n d e r s c h r i j v e n :
ALGEMEEN
Om zinvol te kunnen praten over immunologie is het noodzakelijk om eerst iets te vertellen over c e l l e n , DMA, RNA, a m i n o z u r e n , eiwitten etc. Iedereen heeft deze termen vast wel eens ergens g e h o o r d , maar de betekenis en o n d e r l i n g e samenhang zal niet iedereen duidelijk zijn.
CELLEN
Een cel kunnen we omschrijven als de kleinste eenheid binnen een o r g a n i s m e die nog tot alle b a s i s f u n c t i e s in staat is. Als b a s i s f u n c t i e beschouwen we het in leven blijven en het in staat zijn om zich te
delen. Omdat definities nooit zo erg duidelijk zijn (definitie van een s t o e l ? ) volgen hier enkele v o o r b e e l d e n .
Zoals een huis is o p g e b o u w d uit s t e n e n , een r e k e n m a c h i e n t j e uit c h i p s , zo zijn dieren en planten opgebouwd uit cellen. Hogere dieren en planten zijn opgebouwd uit zeer veel cellen (de mens b.v. uit 2 à 3 0 0 . 0 0 0 . 0 0 0 . 0 0 0 . 0 0 0 c e l l e n ) . Uit de definitie van een cel volgt dat er ook organismen kunnen voorkomen bestaande uit één enkele cel. Voorbeelden hiervan zijn bacteriën en het pan t o f f e l d i e r t j e . Het pan-toffeldiertje heeft het vermogen tot b e w e g e n , voedsel- en zuurstof-opname verenigd in die ene c e l . De cellen van hogere organismen zijn g e s p e c i a l i s e e r d , dat wil zeggen elke cel heeft z'n eigen functie.

Een cel moet aan een aantal vereisten voldoen om in leven te kunnen blijven en om zijn speciale functie uit te kunnen o e f e n e n . Deze ver-eisten zullen we bespreken aan de hand van een voorbeeld: de plasma c e l . De functie van een plasmacel is het produceren van a n t i l i c h a m e n of antistoffen (daar komen we later nog uitvoerig op t e r u g ) . Binnen een cel kunnen we verschillende o n d e r a f d e l i n g e n o n d e r s c h e i d e n : een in-f o r m a t i e c e n t r u m (de k e r n ) , e n e r g i e c e n t r a l e s ( m i t o c h o n d r i ë n ) , in-fabrieken ( r i b o s o m e n ) , a f v a l v e r w e r k i n g s b e d r i j v e n ( l y s o s o m e n ) , i n p a k a f d e l i n g e n ( G o l g i - a p p a r a a t ) en verder nog een i n f r a s t r u c t u u r (endoplasmaties reti culurn). Om te voorkomen dat deze processen in de soep lopen (de a f v a l v e r w e r -king gaat het i n f o r m a t i e c e n t r u m o p r u i m e n ) zijn al deze o n d e r a f d e l i n g e n van elkaar gescheiden door een t u s s e n w a n d j e ( m e m b r a a n ) . De cel als geheel is ook omgeven door een w a n d , de c e l m e m b r a a n . Deze celmembraan zorgt er niet alleen voor dat het hele zaakje bij elkaar gehouden w o r d t , maar ook dat grondstoffen (voor de fabrieken en de e n e r g i e
-c e n t r a l e ) naar binnen of naar buiten kunnen.
Een o r g a n i s m e o n t s t a a t door versmelting van een eicel met een zaad-c e l . Door deling van deze ene zaad-cel ontstaan dan 2 zaad-c e l l e n , die zizaad-ch w e e r d e l e n , e n z . Dat dit snel kan aantikken is bekend uit de parabel van de graankorrel en het schaakbord (fig. 2 ) . W a n n e e r we links bovenaan met 1 graankorrel b e g i n n e n , op het 2e vakje 2 k o r r e l s , op het 3e vakje 4 , e n z . , bereiken we vrij snel de wereld graan p r o d u c t i e . In fig. 2 zien we ook waar we uitkomen voor een volwassen mens als we links boven op het schaakbord beginnen met een eicel en een z a a d c e l .
Bij deling w o r d t de erfelijke informatie (opgeslagen in de kern) keurig verdeeld over de twee cellen. H i e r b i j gaat geen informatie v e r l o r e n , w a n t voordat een cel zich d e e l t , w o r d t eerst alle informatie gecopieerd zodat de twee nieuwe cellen weer de o o r s p r o n k e l i j k e hoe-veelheid b e z i t t e n . Het gevolg hiervan is dat elke cel dus alle e r f e l i j k e informatie b e z i t .

De erfelijke informatie in de kern is gecodeerd. We kunnen dit ver-gelijken met een computer, waar de informatie op een magneetband (DNA) gecodeerd ligt. Als er een bepaalde opdracht moet worden uitgevoerd, b.v. de produktie van anti 1ichamen, dan wordt in de kern het benodigde stukje informatie overgeschreven op "boodschapper RNA". Deze boodschap-per brengt de opdracht (in code) over naar de ribosomen, die zich
buiten de kern verspreid in de cel bevinden. Er zijn verschillende

Zo zijn er losse ribosomen die e i w i t t e n maken voor eigen g e b r u i k , zoals b.v. e i w i t t e n die nodig zijn voor de ver-vanging van versleten o n d e r d e l e n . R i b o s o m e n die e i w i t t e n maken die bestemd zijn voor de e x p o r t (zoals b.v. anti 1 i c h a m e n ) liggen op het e n d o p l a s m a t i e s reticulum zodat de eiwitten meteen naar de i n p a k a f d e -lingen g e t r a n s p o r t e e r d kunnen w o r d e n . E n d o p l a s m a t i e s reticulum met r i b o s o m e n erop noemt men g r a n u l a i r e n d o p l a s m a t i s c h reticulum ( G E R ) ; k a n t - e n - k l a r e e i w i t t e n w o r d e n door het G o l g i - a p p a r a a t ingepakt in b l a a s j e s . Door middel van deze b l a a s j e s w o r d e n de e i w i t t e n uitgeschei-den (fig. 3 ) .
Eiwitten zijn o p g e b o u w d uit a m i n o z u r e n . Er zijn 20 v e r s c h i l l e n d e a m i n o z u r e n . De e i g e n s c h a p p e n van een e i w i t w o r d e n b e p a a l d door het a a n t a l , de s a m e n s t e l l i n g en de volgorde van de a m i n o z u r e n . We kunnen e i w i t t e n v e r g e l i j k e n met w o o r d e n . W o o r d e n zijn o p g e b o u w d uit l e t t e r s . Er zijn 26 v e r s c h i l l e n d e l e t t e r s . De b e t e k e n i s van een woord w o r d t b e p a a l d door het a a n t a l , de s a m e n s t e l l i n g en de volgorde van de let-ters .
I M M U N O L O G I E
Alle gewervelde dieren zijn uitgerust met een b e w a k i n g s s y s t e e m ( i m m u u n s y s t e e m ) , dat er voor zorgt dat b i n n e n g e d r o n g e n vreemde stof-fen of cellen onschadelijk gemaakt w o r d e n . Deze vreemde zaken kunnen b i j v o o r b e e l d bacteriën zijn of virussen of e i w i t t e n . Een i m m u u n s y s -teem moet om die o n g e w e n s t e bezoekers (die we antigenen n o e m e n ) te h e r k e n n e n , o n d e r s c h e i d kunnen maken tussen "vreemd" en "eigen". Onder eigen verstaan we de c e l l e n , e i w i t t e n , hormonen e n z . w a a r u i t het lichaam is o p g e b o u w d . Dit vermogen om o n d e r s c h e i d te kunnen maken is erg belangrijk omdat het natuurlijk niet de bedoeling van het immuun-systeem is om het eigen lichaam af te b r e k e n .
Een voorbeeld uit de praktijk kan veel van de immunologie duidelijk m a k e n . Als je een griepje o p l o o p t kun je een paar dagen goed ziek
zijn. Het griepvirus is dan je lichaam b i n n e n g e d r o n g e n en is zich daar naar hartelust aan het v e r m e n i g v u l d i g e n . Na enige tijd ben je echter w e e r g e n e z e n . Wat er zich in de tussentijd heeft afgespeeld is het v o l g e n d e : het i m m u u n s y s t e e m heeft het virus als vreemd herkend en het daarna o n s c h a d e l i j k g e m a a k t .
Antigenen en antistoffen
Vreemde stoffen roepen een bepaalde reactie op w a n n e e r ze bij mens of dier worden i n g e s p o t e n . De vreemde stoffen noemen we met een ver-zamelnaam a n t i g e n e n . Men heeft gevonden dat voor het oproepen van een i m m u n o l o g i s c h e reactie niet een compleet eiwit of een hele cel nodig is. Een groepje van 6-7 aminozuren is al v o l d o e n d e . Zo'n eenheid noemen we antigene d é t e r m i n a n t .
Als reactie op het inspuiten van een antigeen w o r d e n antistoffen g e p r o d u c e e r d . Deze antistoffen binden specifiek met het a n t i g e e n : dat wil zeggen ze reageren alléén met het antigeen waarmee het dier is in-gespoten en nergens anders m e e . S p e c i f i c i t e i t is één van de belang-rijkste kenmerken van een i m m u n o l o g i s c h e reactie.

A n t i s t o f f e n , die ook wel i m m u n o g l o b u l i n e s w o r d e n g e n o e m d , komen voor in het b l o e d , maar ook in de rest van het lichaam. Bij mensen komen verschillende soorten i m m u n o g l o b u l i n e s voor. De b e l a n g r i j k s t e zijn

In figuur 4 staan IgG en IgM s c h e m a t i s c h g e t e k e n d . Het g e a r c e e r d e gedeelte van het i m m u n o g l o b u l i n e b i n d t ( s p e c i f i e k ) aan het a n t i g e e n . We zien dat IgG 2 b i n d i n g s p l a a t s e n h e e f t , IgM zelfs 1 0 .

Dit alleen is echter niet v o l d o e n d e om b.v. een z i e k t e v e r -w e k k e n d e b a c t e r i e o n s c h a d e l i j k te m a k e n . Dit o n s c h a d e l i j k maken kan op twee m a n i e r e n g e b e u r e n : de e e r s t e m a n i e r is door m a c r o f a g e n (fig.5) Deze holle bolle Gijzen zijn grote cellen die graag rommel o p r u i m e n . Ze doen dat niet s p e c i f i e k ; alles w a t vreemd is wordt o p g e p e u z e l d (macrofaag b e t e k e n t letterlijk v e e l v r a a t ) ,
Omdat immunoglobulines meerdere b i n d i n g s p l a a t s e n h e b b e n , zijn ze in staat om antigenen aan elkaar te doen klonteren. Deze s a m e n g e k l o n t e r d e antigenen zijn dan extra lekker voor m a c r o f a g e n . Een tweede manier waarop antigenen onschadelijk gemaakt worden nadat antilichamen ge-bonden zijn is de volgende.
In het bloed bevindt zich een hele reeks van stoffen die we met een verzamelnaam het complementsysteem noemen. Wanneer een antilichaam gebonden is aan een antigeen (fig. 6) verandert er iets aan het anti-lichaam w a a r d o o r het eerste deel van het c o m p l e m e n t s y s t e e m (Cl) ge-bonden w o r d t . Cl activeert dan C 2 , C3 activeert C4 e n z . totdat het laatste deel C9 geactiveerd w o r d t . C9 is een stof die de membraan van de vreemde cel doet o p l o s s e n , w a a r d o o r de vreemde cel gedood w o r d t .

De belangrijkste kenmerken van het immuunsysteem zijn de s p e c i f i c i t e i t en de vorming van ( i m m u n o l o g i s c h ) geheugen.
Tegen griep kun je je in laten e n t e n . Je krijgt dan een injectie met gedood g r i e p v i r u s . Omdat het virus dood is kan het zich niet meer
ver-m e n i g v u l d i g e n en kun je van zo'n injectie niet ziek w o r d e n . Het
immuunsysteem herkent het griepvirus echter wél als vreemd en reageert d a a r o p . Er worden antistoffen g e v o r m d , maar er w o r d t ook immunologisch geheugen o p g e w e k t . Na een g r i e p s p u i t ben je niet meer vatbaar. Dit b e t e k e n t echter niet dat je dan niet meer met het virus besmet kunt raken. Het virus dringt nog wel het lichaam b i n n e n , maar het immuunsysteem reageert zo snel en zo doeltreffend dat het virus al o n s c h a d e -lijk gemaakt is voordat het je ziek heeft kunnen m a k e n . Je bent dan immuun voor de griep. Het immunologisch g e h e u g e n , dat daarbij gevormd w o r d t , houdt lang aan. De bekende DTKP prikken maken je gedurende de rest van je leven immuun voor d i f t e r i e , t e t a n u s , k i n k h o e s t en p o l i o . Toch biedt een griepspuit over het algemeen maar bescherming voor één j a a r ; het volgend jaar kun je toch weer de griep k r i j g e n . Dit w o r d t
niet veroorzaakt door een kortdurend immunologisch geheugen maar heeft alles te maken met de s p e c i f i c i t e i t van het i m m u u n s y s t e e m . Een griepepidemie krijgt net als orkanen een naam: de A g r i e p , de H o n g k o n g -griep e n z . Men gebruikt die verschillende namen omdat het telkens weer een ander virus b e t r e f t . Alhoewel die verschillende virussen wel alle-maal griep v e r o o r z a k e n , verschillen ze onderling zó sterk dat het im-muunsysteem zich niet meer herinnert daarmee ooit in aanraking te zijn g e w e e s t . Een A-griep spuit biedt dan ook geen b e s c h e r m i n g tegen een infectie met het H o n g k o n g - g r i e p v i r u s .
Humorale en cellulaire immuniteit
In de immunologische reacties die we tot nu toe besproken hebben w o r d t de vreemde stof onschadelijk gemaakt met behulp van a n t i 1 i c h a m e n . Men noemt deze vorm van verdediging humoral i m m u n i t e i t . Humoraal b e t e -kent v l o e i b a a r . Een dergelijke naam is gekozen omdat deze vorm van immuniteit o v e r g e b r a c h t kan worden door een dier met serum van een immuun dier in te s p u i t e n . De cellen die betrokken blijken te zijn bij humorale reacties heten B - l y m f o c y t e n .
Een andere vorm van immuniteit is de zogenaamde cellulaire immuniteit. Voorbeelden uit de praktijk zijn de afstoting van een g e t r a n s -planteerd orgaan en de M a n t o u x - r e a c t i e (krasjes bij TBC-control e ) . Bij een cellulaire immunologische reactie worden geen antilichamen g e m a a k t , de vreemde cel wordt onschadelijk gemaakt door direct contact met een lymfocyt (fig. 7 ) . De lymfocyten betrokken bij cellulaire immuniteit noemt men T - l y m f o c y t e n . De twee kenmerken van een i m m u n o l o g i s c h e reactie s p e c i f i c i t e i t en geheugen gelden voor zowel humorale als cellulaire r e a c t i e s .

Organen
Belangrijke immuunorganen bij de mens zijn m i l t , thymus ( z w e z e r i k ) , b e e n m e r g , lymfeklieren en in mindere mate tonsillen (amandelen) en appendix (blinde d a r m ) . T-lymfocyten komen uit de thymus en B-lymfo-cyten worden aangemaakt in het beenmerg. Vissen bezitten geen beenmerg of lymfeklieren. De belangrijkste organen bij een vis zijn t h y m u s , m i l t , kopnier en nier. (fig. 8 ) .

De thymus bestaat vrijwel uitsluitend uit l y m f o c y t e n , in m i l t , kopnier en nier komen naast lymfocyten ook veel rode bloedcellen en
granulo-cyten voor. Behalve in de immuunorganen komen lymfogranulo-cyten ook voor in het bloed waar ze behoren tot de witte b l o e d l i c h a a m p j e s .
Immunologische reactie
Wat gebeurt er nu precies als een vreemde stof het lichaam is bin-n e bin-n g e d r o bin-n g e bin-n . We hebbebin-n al besprokebin-n dat abin-ntilichamebin-n specifiek eebin-n vreemde stof kunnen binden. Lymfocyten hebben antilichamen aan hun c e l o p p e r v l a k . Door middel van deze oppervlakte-anti 1ichamen zijn ze in staat om vreemde stoffen te h e r k e n n e n . Deze herkenning is specifiek, d.w.z. een lymfocyt herkent maar één bepaalde vreemde stof en een vreemde stof wordt maar door één lymfocyt (of een groepje identieke l y m f o c y t e n ) als vreemd herkend. In figuur 9 is deze specifieke her-kenning vergeleken met een aangerande maagd (lymfocyt) die met haar hand (oppervlakte-anti 1ichaam) uit een hele reeks van verdachten haar aanvaller (vreemde cel) herkent aan de vorm van zijn oren (antigene determi n a n t ) .

Nadat de herkenning heeft plaats gevonden (tussen vreemde stof A en lymfocyt a) gaat de desbetreffende lymfocyt zich delen. Op die manier ontstaan er veel lymfocyten die allemaal specifiek zijn voor de vreem-de stof A. Daarna veranvreem-deren (differentiëren met een mooi w o o r d ) vreem-de
lymfocyten in p l a s m a c e l l e n . Plasmacellen produceren veel anti 1ichamen (specifiek gericht tegen vreemde stof A ) en deze a n t i l i c h a m e n komen onder andere in het bloed t e r e c h t . De manier w a a r o p a n t i l i c h a m e n een vreemde stof herkennen en onschadelijk maken hebben we reeds b e s p r o k e n .
Nadat herkenning heeft p l a a t s g e v o n d e n veranderen gelukkig niet alle lymfocyten in een p l a s m a c e l . Een gedeelte w o r d t namelijk g e h e u g e n c e l . Deze g e h e u g e n c e l l e n zorgen ervoor dat als er weer een infectie met vreemde stof A optreedt het hele proces van herkenning tot en met anti l i c h a a m p r o d u c t i e veel sneller en heftiger v e r l o o p t .
Als voorbeeld dient een grafiek va.n het aantal plasmacellen in de kop-nier van een karper tijdens een eerste en een herhaalde reactie (fig. 1 0 ) .

T E C H N I E K E N
In dit hoofdstukje worden enkele technieken besproken die g e b r u i k t zijn bij het onderzoek van het i m m u u n s y s t e e m van de k a r p e r .
Rozet test
Met behulp van de rozet test is het mogelijk om het aantal lymfo-cyten gericht tegen een bepaalde vreemde stof te b e p a l e n . We maken d a a r b i j gebruik van het feit dat de o p p e r v l a k t e - a n t i 1ichamen van een lymfocyt een vreemde stof kunnen b i n d e n .
Van een karper die ingespoten is met konijne rode b l o e d c e l l e n (KRBC) v e r w i j d e r e n we de m i l t . De milt w o r d t f i j n g e k n i p t en door een fijnmazig gaasje gewreven zodat we een o p l o s s i n g krijgen bestaande uit losse m i l t c e l l e n . Aan deze c e l s u s p e n s i e voegen we KRBC toe en v e r v o l -gens zetten we dit m e n g s e l t j e een tijdje in de k o e l k a s t . Als er in de milt lymfocyten aanwezig waren specifiek gericht tegen K R B C , dan bin-den de o p p e r v l a k t e - a n t i 1ichamen van de l y m f o c y t aan de KRBC. Op die m a n i e r o n t s t a a t er een krans van KRBC rondom zo'n l y m f o c y t : een rozet
(fig. 1 1 ) . Door op v e r s c h i l l e n d e dagen na inspuiten het aantal rozet-ten te tellen kun je een idee krijgen hoe de lymfocyrozet-ten reageren op deze 1 i c h a a m s v r e e m d e stof.

Bij de plaque test bepalen we het aantal p l a s m a c e l l e n dat anti-lichamen uitscheidt tegen een bepaalde vreemde stof of c e l . Als vreem-de cel gebruiken we nu niet KRBC maar schape rovreem-de b l o e d c e l l e n ( S R B C ) . Ook hier maken we w e e r een c e l s u s p e n s i e en mengen deze met SRBC.
Bovendien w o r d t c o m p l e m e n t aan het mengsel t o e g e v o e g d . Dit m e n g s e l t j e laten we tussen twee m i c r o s c o o p glaasjes l o p e n . De SRBC en de karper-cellen zijn zodanig verdund dat 1) tussen de glaasjes een m o n o l a y e r
o n t s t a a t , d . w . z . de cellen liggen tegen elkaar aan in een enkele laag; 2) er veel meer SRBC zijn dan k a r p e r c e l l e n . Als er in h e t mengsel
plasmacellen aanwezig waren dan gaan deze cellen tussen de glaasjes rustig door met de taak waar ze mee bezig w a r e n , namelijk het maken en uitscheiden van anti l i c h a m e n . De a n t i l i c h a m e n binden zich aan de S R B C , die immers in o v e r m a a t aanwezig z i j n , vervolgens w o r d t h e t com-p l e m e n t geactiveerd en de SRBC in de buurt van de com-plasmacel worden ge-dood. Op die m a n i e r o n t s t a a t er rondom de plasmacel een gat dat na verloop van tijd met het blote oog kan worden w a a r g e n o m e n . Door het aantal gaten (plaques) te tellen komen we te weten hoeveel p l a s m a -cellen er aanwezig waren in een bepaald o r g a a n . Foto's van plaques staan in appendix I en I I .
S c h u b t r a n s p l a n t a t i e
Een methode om de cellulaire i m m u n i t e i t te testen is door schubben te t r a n s p l a n t e r e n en vervolgens te bekijken hoe lang het duurt voordat zo'n schub afgestoten i s .
Bij de karper liggen de schubben als dakpannen over e l k a a r heen. Het is mogelijk om bij een vis een schub te verwijderen en daarvoor een schub van een andere vis in de plaats te z e t t e n . In figuur 12 staat aangegeven hoe we dat ongeveer gedaan h e b b e n .

Een getransplanteerde schub w o r d t herkend als 1ichaamsvreemd en het immuunsysteen neemt daarom m a a t r e g e l e n : de p i g m e n t c e l l e n , die in een normale schub rond zijn (fig. 1 3 ) , vertakken zich. Wat later krijgt de getransplanteerde schub een meikwit-achtige schijn over zich. Dit wordt veroorzaakt door de lymfocyten die op de vreemde schub afkomen en deze afbreken. Na een aantal dagen verdwijnen de lymfocyten weer en blijft er alleen nog maar een doorschijnend stukje bot over. Op dit moment beschouwen we de schub als afgesoten.
IMMUNOLOGIE VAN DE KARPER
In het voorafgaande is de karper al af en toe ter sprake gekomen in verband met het immuunsysteem. In appendix I van dit proefschrift staan de resultaten vermeld van onderzoek verricht aan een tropisch visje Barbus aonchonius (prachtbarbeel in het n e d e r l a n d s ) . Aandacht werd besteed aan de ontwikkeling van het humorale en cellulaire immuun-systeem. Humorale immuniteit werd onderzocht door bij dieren van ver-schillende leeftijden te kijken naar het aantal plaque vormende cel-len dat in de milt ontstaat na inspuiten met SRBC. Cellulaire immuni-teit werd onderzocht met schubtransplantaties. De resultaten laten zien dat dieren van 3 maanden al kunnen reageren op S R B C , maar dat het daarna nog wel 6 maanden duurt voordat dit vermogen maximaal is. De cellulaire immuniteit ontwikkelt zich sneller: dieren van 6 maanden oud stoten vreemde schubben even snel af als 9 maanden oude dieren.
In appendix II staat hoe we de plaque test bij de karper uitgevoerd hebben. Het grootste probleem was het vinden van een geschikte com-plement bron. Het bleek dat karper-comcom-plement niet het meest geschikt w a s . Veel beter was kopvoorn, sneep, barbeel en brasem. Omdat de kop-voorn, sneep en barbeel in Nederland erg zeldzaam of zelfs beschermd zijn, hebben we besloten de optimale omstandigheden uit te werken voor brasem-complement. Brasems zijn er gelukkig genoeg, er is zelfs een meer naar genoemd.
In appendix III is gekeken naar de reactie van het immuunsysteem van de karper op SRBC bij verschillende temperaturen. Eerst hebben we gekeken in welke organen antilichaam producerende cellen voorkomen. In de kopnier en nier zit ongeveer 90% van het totaal aantal "plaque
vormende cellen" ( P F C ) . Slechts 5% zit in de milt en verder komen er nog geringe aantallen PFC voor in het bloed, het hart en de thymus. In figuur 10 staat weergegeven hoe het aantal PFC in de kopnier ver-loopt bij 24 C. Het hoogste aantal PFC wordt bereikt op dag 9 na
in-spuiten. Als we de karpers bij een lagere temperatuur houden, dan duurt het langer voordat de piek van de PFC reactie bereikt w o r d t .
Vissen zijn koudbloedige dieren, ze hebben geen vaste lichaamstempera-tuur zoals zoogdieren maar de temperalichaamstempera-tuur van hun lichaam wordt bepaald door de omgevingstemperatuur. Bij lagere temperaturen verlopen alle levensprocessen van een vis trager. Het is daarom niet verwonderlijk dat ook de immuunreactie trager verloopt. Wat echter opviel was dat de hoogte van de reactie (het maximale aantal antilichamen vormende cellen) gelijk was bij alle temperaturen. Van uitstel komt in dit ge-val dus geen afstel. Een tweede interressant aspect van dit experiment blijkt als je in een grafiek de temperatuur uitzet tegen de dag waarop
de p i e k r e a c t i e b e r e i k t w o r d t ( f i g . 1 4 ) . We zien dat de g r a f i e k geen rechte lijn v o r m t m a a r dat er een " k n i k " in z i t .

In a p p e n d i x IV staan e x p e r i m e n t e n b e s c h r e v e n die gedaan zijn om iets te w e t e n te komen o v e r v o r m i n g van i m m u n o l o g i s c h g e h e u g e n . We h e b b e n al b e s p r o k e n dat als je een d i e r met een v r e e m d e s t o f i n s p u i t e r niet u i t s l u i t e n d s p e c i f i e k e a n t i l i c h a m e n g e v o r m d w o r d e n m a a r ook
g e h e u g e n c e l l e n . De g e h e u g e n c e l l e n zorgen e r v o o r dat na een tweede in-j e c t i e met d e z e l f d e v r e e m d e s t o f de r e a c t i e s n e l l e r en f e l l e r i s . De m a t e van g e h e u g e n v o r m i n g kun je dus a f l e z e n aan de h o o g t e van de h e r -h a a l d e r e a c t i e .
Karpers zijn i n g e s p o t e n met 3 v e r s c h i l l e n d e h o e v e e l h e d e n SRBC (1 m i l j a r d , 10 m i l j o e n en 1 0 0 . 0 0 0 c e l l e n / d i e r ) en l a n g s 2 v e r s c h i l l e n d e r o u t e s : in de s p i e r e n of in de b l o e d b a a n . De h o o g s t e dosis SRBC g e e f t de h o o g s t e e e r s t e reactie terwijl b i j de l a a g s t e dosis n a u w e l i j k s een reactie te meten v a l t . Als je e c h t e r na 1 m a a n d de g e h e u g e n v o r m i n g test d o o r een tweede i n j e c t i e toe te dienen dan b l i j k t dat de b e s t e r e a c t i e w o r d t g e v o n d e n in dieren die met de m i d d e l s t e dosis g e i m m u n i -seerd w o r d e n . Na 6 m a a n d e n en 1 j a a r is de s i t u a t i e nog e x t r e m e r : de h o o g s t e r e a c t i e w o r d t b e r e i k t in d i e r e n g e i m m u n i s e e r d met de l a a g s t e d o s i s . Dat b e t e k e n t dat de d o s i s die in een e e r s t e r e a c t i e n a u w e l i j k s iets d o e t , het b e s t e is voor de geheugen v o r m i n g . Je kunt dit f e n o m e e n ook v e r t a l e n in termen van v a c c i n e r e n ( i n e n t e n ) en b e s c h e r m i n g tegen een z i e k t e ( i m m u n i t e i t ) . Het idee h e e r s t nog steeds dat een vaccin
een zo hoog m o g e l i j k e anti 1 i c h a a m p r o d u c t i e op moet wekken wil het bescherming b i e d e n . Op grond van deze e x p e r i m e n t e n zou je het t e g e n o v e r -gestelde kunnen b e w e r e n : een vaccin b i e d t pas dan o p t i m a l e b e s c h e r m i n g als het bij een eerste injectie geen a n t i l i c h a m e n o p w e k t .
In de laatste twee h o o f d s t u k k e n , appendix V en V I , w o r d t beschreven wat voor effecten antibiotica op het i m m u u n s y s t e e m van de karper kun-nen h e b b e n .
A n t i b i o t i c a zijn stoffen die de groei van bacteriën remmen. De b e -kendste zijn p e n i c i l l i n e , s t r e p t o m y c i n e , c h l o o r a m p h e n i c o l , tetra-c y tetra-c l i n e . A n t i b i o t i tetra-c a worden bij de mens gebruikt voor de b e s t r i j d i n g van i n f e c t i e z i e k t e s . In de v e e h o u d e r i j worden ze behalve voor de be-strijding van ziektes ook p r e v e n t i e f (ter voorkoming van z i e k t e s ) ge-b r u i k t . Een ge-b i j k o m e n d e reden voor het gege-bruik is de groei ge-b e v o r d e r e n d e werking op de d i e r e n . Het w e r k i n g s m e c h a n i s m e is niet voor alle a n t i -biotica gelijk. P e n i c i l l i n e b.v. v e r s t o o r t de vorming van een goede c e l w a n d , w a a r d o o r de b a c t e r i e , als hij gaat g r o e i e n , uit e l k a a r k l a p t . Dit kunnen we vergelijken met de gevolgen van een kapotte b u i t e n b a n d van een f i e t s ; de b i n n e n b a n d klapt pas als je hem o p p o m p t . Tetra-cycline en c h l o o r a m p h e n i c o l remmen de e i w i t f a b r i e k van b a c t e r i ë n .
A n t i b i o t i c a remmen de groei van b a c t e r i ë n . Alleen remming van de groei is natuurlijk niet voldoende om een bacterie uit te s c h a k e l e n , immers zodra je stopt met de b e h a n d e l i n g gaan de bacteriën w e e r rustig verder met groei en deling. Daarom zal altijd het i m m u u n s y s t e e m nodig zijn om d e , door antibiotica in bedwang gehouden b a c t e r i ë n , d e f i n i t i e f onschadelijk te m a k e n .
Het a n t i b i o t i c u m dat we gebruikt hebben in onze e x p e r i m e n t e n is Oxy-tetracycline ( o x y T C ) . De s c h e i k u n d i g e formule staat in figuur 15.

Het a n t i b i o t i c u m is op twee v e r s c h i l l e n d e manieren toegediend aan de karper.
1. door karpers te voeren met korrels waarin oxyTC m e e g e m e n g d i s . Dit is ook de manier w a a r o p antibiotica in de v i s t e e l t worden toe-gediend.
2. door karpers om de 3 dagen met een o x y T C - o p l o s s i n g in te s p u i t e n . Bij de b e h a n d e l d e karpers hebben we gekeken of het i m m u u n s y s t e e m nog goed w e r k t e . Het bleek dat het bij k a r p e r s , die met oxyTC waren i n g e -s p o t e n , veel langer duurde voordat g e t r a n -s p l a n t e e r d e -schubben w e r d e n afgestoten dan bij c o n t r o l e - d i e r e n . Er waren zelfs schubben die hele-maal niet meer werden a f g e s t o t e n . Injecties met oyxTC remmen dus in sterke mate de cellulaire i m m u u n r e a c t i e s . De humorale i m m u n i t e i t wordt sterk geremd door zowel injecties als voeren van o x y T C :

Zowel het aantal rozet vormende cellen als het aantal a n t i l i c h a a m producerende cellen is sterk verminderd tijdens een humorale respons in met antibiotica b e h a n d e l d e d i e r e n .
In latere e x p e r i m e n t e n hebben we dit effect op de humorale i m m u n i -teit verder o n d e r z o c h t . Daarbij bleek dat vooral de eerste reactie gevoelig voor antibiotica w a s , herhaalde reacties werden niet beïn-vloed .
Uit deze e x p e r i m e n t e n kun je concluderen dat er erg v o o r z i c h t i g met a n t i b i o t i c a moet worden o m g e s p r o n g e n . I m m e r s , w a n n e e r je een ziek d i e r , dat een bepaalde infectie h e e f t , met a n t i b i o t i c a gaat b e h a n d e l e n dan gebeuren er twee dingen:
a. de bacterie wordt door het a n t i b i o t i c u m in zijn groei geremd b. het i m m u u n s y s t e e m van het dier wordt o n d e r d r u k t .
Als het dier tijdens de a n t i b i o t i c u m k u u r besmet w o r d t met een v i r u s , een schimmel of een bacterie die niet gevoelig is voor het a n t i b i o t i cum dan kan het i m m u u n s y s t e e m deze m i c r o o r g a n i s m e n niet meer o n s c h a -delijk m a k e n . Tenslotte is het de vraag wie zich na beëindiging van de a n t i b i o t i c u m b e h a n d e l i n g het snelst h e r s t e l t : de bacterie of het im-m u u n s y s t e e im-m .

OPENING REMARKS
There are several reasons for studying the immune system of f i s h :
1) From a phylogenetic point of view fish are i n t e r e s t i n g because they are the f i r s t group of animals in which an immune system characterized by the presence of immunoglobulins occurs.
2) The immune system of poikilotherms - including fish - i s dependent upon the environmental temperature offering the unique p o s s i b i l i t y to manipulate the immune response by a mere v a r i a t i o n i n temperature.
3) When studying the ontogeny of the immune response oviparous fish are p a r t i c u l a r i n t e r e s t i n g because the e a r l y stages and free swimming larvae are r e a d i l y accessible to experimental work.
4) A b e t t e r understanding of the immune system of fish may aid i n prophylaxis and therapy of fish diseases in fish c u l t u r e .
In the General Introduction relevant data about fish immunology w i l l be discussed. Since i t i s not the i n t e n t i o n of t h i s t h e s i s to review the whole f i e l d of comparative immunology, data are r e s t r i c t e d to fishes although a comparison with i n v e r t e b r a t e defence mechanisms and mammalian immune systems w i l l be made when needed.
During the l a s t 5-10 years the i n t e r e s t in fish immunology has accumulated in a vast number of p u b l i c a t i o n s . However the data are d i f f i c u l t to unify: only in the introduction of t h i s t h e s i s 69 species are mentioned. During evolution the d i f f e r e n t classes of fish have diverged long before mammals were p r e s e n t . For t h i s reason i t i s d i f f i c u l t , i f not impossible, to draw d e f i n i t e conclusions when comparing data of species belonging to different c l a s s e s . In t h i s respect i t i s worthwhile t o mention the observation of Heuzeroth, Resch, Richter & Ambrosius (1973) who studied the degree of s i m i l a r i t y of immunoglobulins. The differences between c l a s s e s of fishes were as large as the differences between other v e r t e b r a t e orders ( e . g . amphibians and marnais). Even within one class of fish data are d i f f i c u l t to compare due to a v a r i e t y of immunization schedules and antigens used. Moreover, species may d i f f e r in t h e i r p r e f e r e n t i a l temperature. In the Epilogue an attempt w i l l be made to give a general and more personal idea about the immune system of bony f i s h .
In most experiments described i n t h i s t h e s i s carp (Cyprinns oarpio L. 1758) were used. This animal was chosen because i t i s an excellent experimental animal for b i o l o g i c a l s t u d i e s . In addition i t i s worthwhile to mention t h a t t h i s species,Tilapia and salmonids form the base for large scale fish c u l t u r e in the world.
G E N E R A L INTRODUCTION
CHAPTER I CELLS AND ORGANS
Cells
At the light microscopical as well as electron-microscopical level lymphocytes, plasma cells, mononuclear phagocytes and granulocytes of fish closely resemble their mammalian counterparts.
Lymphocytes are small round cells with a large nucleus and a small rim of basophilic cytoplasm (Weinreb, 1963; Ferguson, 1976b; Mattison & Fänge, 1977; Kreuzmann, 1977; Davina, Rijkers, Rombout, Timmermans & van Muiswinkel, 1980).
Plasma cells possessing an eccentric nucleus with a prominent nucleolus and a cytoplasm packed with rough endoplasmic reticulum have been observed in rainbow trout (Chiller, Hodgins, Chambers & Weiser, 1969; Etlinger, Hodgins & Chiller, 1978).
Monocytes and macrophages have been described in fish but a confusing nomen-clature is used by many authors (see review of Ellis, 1977a). The same holds true for granulocyte identification. A description of the morphology and a discussion on the nomenclature of these cells is published by Barber & Westermann (1975, 1978), Ellis (1977a) and Davina et al. (1980).
©Lymphoid cells as well as mononuclear phagocytes and granulocytes of mammals and fish are quite comparable when morphological criteria are used.
Lymphoid organs
The most primitive vertebrates, the hagfish lack on morphological grounds a definite thymus and spleen (Harboe, 1963). Haematopoietic foci are present in the lamina propria of the entire gut lenght (Good, Finstad, Pollara & Gabrielsen,
1966). The pronephros, which contains nephrostomes, is transformed partly into lymph-oid tissue (Gérard, 1954; Fänge, 1966). In peripheral blood of sea lamprey, cells were observed which resembled mammalian lymphocytes. A primitive spleen is located in an invagination of anterior gut tissue. Erythro-, thrombo-, granulo- and lympho-poiesis occurs in this organ. A primitive bone marrow, located in the fibrocartilagi-nous prevertebral arch, has been observed. The bone marrow contains haematopoietic tissue; proliferation of lymphoid cells was observed after stimulation with bovine gamma globulin (BGG). A definite thymus is absent in lamprey. However, small groups of lymphoid cells in the epithelium of the pharyngeal gutter are considered as a primitive thymus (Good et al., 1966).
From the evolutionary point of view, the guitarfish (a primitive Elasmobranch) is the first representative where a thymus is found. It is a fully developed,
encapsulated lymphoid organ with a well organized cortex and medulla. In the spleen a red and white pulp area is found. Furthermore lymphoid tissue was found in the gut and in the gonads. In the more advanced Elasmobranchs, the leopard shark and nurse shark, a well developed thymus and spleen were present. Abundant lymphoid tissue was also found along the gastrointestinal tract and in the gonads. The kidney did not contain lymphoid cells. Cells, identical to mammalian plasma cells, were found among the lymphoid cells in spleen and gonads (Good et al., 1966).
The Chondrostean paddlefish has a well-developed thymus organized into lobules. Peripheral blood contains large, medium and small lymphocytes. The spleen is well developed and clearly divided into red and white pulp. Plasma cells were observed in spleen and in haematopoietic tissue overlying the heart. The holostean bowfin has a thymus with some degree of organization into cortex and medulla. The spleen is a discrete organ with tissue components distributed into red and white pulp
(Good et al., 1966). In holostean fish a unique organ is found, the meningeal myeloid tissue, which is actively involved in haemopoiesis and thought to be primitive bone marrow (Scharrer, 1944; McLeod, Sigel & Yunis, 1978).
In teleost fish, no uniformity exists in the histological appearance of lymphoid organs. For instance the thymus of eel (von Hagen, 1936) and Mozambique mouthbrooder (Sailendri & Muthukkaruppan, 1975) is clearly divided into a cortex and a medulla whereas this distinction can hardly be made in the salmon and rainbow trout (Ellis, 1977b; Grace & Manning, 1980). A spleen with red and white pulp is
found in Mozambique mouthbrooder (Sailendri & Muthukkaruppan, 1975), perch (Pontius & Ambrosius, 1972) and pike (M.G. Vos, pers. comm.). In plaice this division is less clear, the area occupied by the white pulp being relatively small (Ellis & de Sousa, 1974), whereas in carp (Secombes & Manning, 1980) and rainbow trout, spleen is mainly red pulp (Anderson, 1974; Grace & Manning, 1980). The kidney is an important lymphoid organ in teleosts. The excretion function of pronephros is completely lost in adult fish and the organ shows mainly haemopoietic and lymphoid cells (Ellis & de Sousa 1974). Some sinuses in pronephros are surrounded by endocrine cells (Secombes & Manning, 1980). Lymphoid tissue of mesonephros is situated in between nephric tu-bules. In Mozambique mouthbrooder no organized lymphoid tissue was observed in mesonephros (Sailendri & Muthukkaruppan, 1975). Furthermore individual or small groups of lymphoid cells are situated in between intestinal epithelial cells as well as in the lamina propria (Bullock, 1963; Pontius & Ambrosius, 1972; Krementz & Chapman, 1975; Reiffel & Travill, 1977; Zapata, 1979a; Davina et al., 1980). Tele-ost fish lack bone marrow, bursa of Fabricius and lymph nodes.
# I n cartilaginous and bony fish a discrete thymus and spleen are found. In bony fish the kidney contains lymphoid cells. With the exception of sea lamprey and Holosteans, fish lack bone marrow. The cellular organization of thymus and spleen is strongly species dependent.

After injection of carp with a soluble antigen (human gamma globulin (HGG)), antigen trapping occurred in spleen, pronephros and to a lesser degree in mesonephros. Using a cellular antigen (Aeromonas salmonieida), mesonephros played a major part in antigen trapping. Thymus and liver were not involved in this proces (Secombes & Manning, 1980). Antigen binding cells (or rosette forming cells) were detected in spleen and pronephros of rainbow trout after immunization with sheep red blood cells
(SRBC) (Chiller, Hodgins, Chambers & Weiser, 1969). Unfortunately mesonephros was not included in this study. Electron microscopy revealed that lymphocytes, blast-like cells, macrophages and cells resembling eosinophils bound the antigen. Antigen binding cells have also been observed in pronephros and spleen of goldfish, but only low numbers were found in thymus (Ruben, Warr, Decker & Marchalonis, 1977; Warr, DeLuca, Decker, Marchalonis & Ruben, 1977). Antibody producing cells have
been demonstrated in spleen and pronephros of bluegill (Smith, Potter & Merchant, 1967), rainbow trout (Chiller, Hodgins & Weiser, 1969; Anderson, 1978), perch, (Pontius & Ambrosius, 1972) and Mozambique mouthbrooder (Sailendri & Muthukkaruppan,
1975). Furthermore antibody forming cells were detected in the mesonephros of rainbow trout and goldfish (Anderson, 1978; Neale & Chavin, 1971). Organ cultures of snapper and grouper revealed that antibody synthesis occurred not only in spleen and pronephros but also in the thymus (Ortiz-Muniz & Sigel, 1968, 1971). In Mozam-bique mouthbrooder antibody forming cells were present in the thymus (Sailendri,
1973). In other teleost species studied, thymus contained no or very low numbers of antibody forming cells. In liver of rainbow trout and several shark species antibody forming cells were detected (Chiller, Hodgins & Weiser, 1969; Gitlin, Perricelli & Gitlin, 1973).
Ontogenetic studies in salmon (Ellis, 1977b) and carp (Grace & Manning, 1980), suggested that the spleen is not vital for immunological maturity since lymphocytes of thymus and kidney carry surface immunoglobulin and display mixed leucocyte re-actions at a time when spleen in only present in a rudimentary form. Splenectomy in killifish did not affect allograft rejection (Goss, 1961). In snapper, splenectomy had no effect on the antibody response against bovine serum albumin (BSA)(Ferren,
1967). On the other hand, the spleen of blue gourami is supposed to be a major lymphoid organ (Yu, Sarot, Filazzola & Perlmutter, 1970).
Adult thymectomy in Mozambique moutbrooder and salmon had no effect upon allo-graft rejection (Sailendri, 1973; Botham, Grace & Manning, 1980). Thymectomy in 4 months old Mozambique mouthbrooder prolonged survival of allografts and thymectomy in 2 months old animals totally suppressed the anti-SRBC response (Sailendri, 1973). The pronephros has been regarded as a phylogenetic analogue of bone marrow (Zapata,
1979b) and/or lymph node (Ellis, 1977a).
# T h e thymus of bony fish may be looked upon as a primary lymphoid organ important for the continuous production of lymphoid cells. Kidney (pronephros and meso-nephros) can be considered as a stem cell compartment, a primary lymphoid organ
and a peripheral lymphoid organ. It seems that the spleen does not play an important role in the immune response of most bony fish.
CHAPTER 2
NON-LYMPHOID DEFENCE MECHANISMS
Non-lymphoid cellular defence
Phagocytosis of foreign material both serves as a defence mechanism in itself and as an initial step in the onset of the specific immune response. In mammals, mononuclear phagocytes as well as cells belonging to the granulocyte series posses phagocytic capacity.
In the holostean gar studies in vivo and in vitro showed that monocytes and macrophages were capable to phagocytize bacteria, yeast and SRBC. These cells des-troyed the engulfed micro-organisms, as revealed by electron microscopy. Granulocytes appeared to be inert in vivo and in vitro uptake of particles (McKinney, Smith,
Haines & Sigel, 1977). In plaice, monocytes and thrombocytes were the only phagocytic cell types in peripheral blood (Ellis, 1976; Ferguson, 1976b). Macrophages found in pronephros, mesonephros, spleen, thymus, heart, mesentary and peritoneal fluid, consisted of three types: 1) free rounded cells resembling monocytes 2) reticulo-endothelial (RE) cells lining blood sinuses and 3) melano-macrophages. After intra-peritoneal injection of carbon particles, phagocytosis was performed predominantly by the ellipsoids of the spleen and the network of RE cells throughout haemapoietic
tissue of pronephros and mesonephros (Ellis, Munroe & Roberts, 1976). Macrophages lining the atrial endocardium were also involved in phagocytosis (Ferguson, 1975). Following phagocytosis some macrophages in pro- and mesonephros and spleen formed aggregates with melano-macrophages (Ellis et al., 1976, M.G. Vos, pers. comm.). Amazingly, granulocytes were not phagocytic in plaice (Ellis, 1976).
In contrast to the situation in gar and plaice, neutrophilic granulocytes in rainbow trout and goldfish have phagocytic properties (Finn & Nielsen, 1971a, 1971b; Watson, Shechmeister & Jackson, 1963; Weinreb & Weinreb, 1969). Eosinophilic granulocytes of goldfish phagocytize bacteria (Watson et al., 1963; Weinreb & Weinreb, 1969). Similar observations have been made in the cunner (Mackmull & Michels, 1932), carp (Pliszka, 1939) and guppy (Jakowska & Nigrelli, 1953).
Thrombocytes have been described to be phagocytic (Yokoyama, 1960; Fänge, 1968; Ferguson, 1976b). It remains to be demonstrated for these cells if the uptake of
foreign material is followed by intracellular digestion.
There are a number of publications dealing with natural or experimental infec-tions in fish. An extensive discussion of these reports falls beyond the scope of this thesis. In this context it is only worthwhile to mention that phagocytic cells are involved in inflammatory reactions evoked by viruses (Finn, 1970), bacteria Post, 1963) and parasitic infections (Joy & Jones, 1973).
In some studies the clearance of carbon or antigenic material from blood was used as an indication for the phagocytizing capacity of the animal. Following intra-venous or intramuscular injection of T2 bacteriophage into lemon shark, phage
Evans, Russell, Wells, Painter & Lucas, 1968). In brown trout and carp, kept at opti-mal temperatures MS2 bacteriophage is cleared from the bloodstream within 7 days. In the icefish, kept at 2°C this process takes 42-56 days (O'Neill, 1980). The effect of temperature on non lymphoid defence will be discussed in Chapter 7.
• Phagocytosis in fish is accomplished by mononuclear phagocytes. In some species granulocytes are also involved in this process.
Non-lymphoid humoral defence
In higher vertebrates a number of components have been described with a potent anti-bacterial and/or anti-viral activity which function in a non-specific way. Although some components act totally independent of the immune system others are activated by antibody (e.g. complement).
The following components present in higher vertebrates have been described for fish. Their biological significance and their relation to the specific defence sys-tem, especially under circumstances were the immune system functions poorly (low temperatures), are largely unknown at present.
a) complement
Complement is a group of serum components involved in both specific and non-specific defence. In mammals the complement system consists of a series of at least 18 proteins (including C1-C9) which can be activated in two ways. In the classical pathway complement is activated by contact with the altered Fc part of an antibody molecule after binding with an antigen. In the alternative pathway activation is
accomplished by contact with bacterial cell wall polysaccharides. Complement activity has been demonstrated in sera from all vertebrate classes (Gewürz, Eugster, Muschel, Finstad & Good, 1965; Gewürz, Finstad, Muschel & Good, 1966; Legier, Evans & Dupree, 1966; Legier, Evans & Dupree, 1967; Gigli & Austin, 1971; Ross & Jensen, 1973). Fish sera in which complement activity has been demonstrated are shown in TABLE 1.
Basic properties of mammalian complement (thermolability, requirement of Ca , Mg ) are shared by fish complement. However, the temperature range over which com-plement remains active is far greater for fishes: at 4 C perch and carp comcom-plement retains its haemolytic activity (Pontius & Ambrosius, 1972; own observations). For heat-inactivation of fish complement lower temperatures are required than to inactivate mammalian complement (TABLE 1).
As in other poikilotherms complement is in most cases not exchangable
between unrelated fish species. SRBC, sensitized by rainbow trout antibodies, were haemolyzed only in combination with isologous serum (rainbouw trout) or with serum from closely related species as coho salmon, sockeye salmon and chinook salmon
(Chiller, Hodgins & Weiser, 1966). For SRBC, sensitized with blue gill antibodies, blue gill, pumpkin seed and white perch serum are effective complement sources
(Smith, Potter & Merchant, 1967). Rosy barb and rainbow trout serum are suitable complement sources for SRBC sensitized with rosy barb antibodies (Rijkers &

In the haemolytic plaque assay using SRBC and immune carp cells, sera of carp, bream, barbel, chub and nase were capable of bringing about haemolysis (Rijkers, Frederix-WoIters & Van Muiswinkel, 1980a). It appears that isologous serum or serum from closely related species is effective as complement source in the haemolytic plaque assay. However, in the plaque assay of rosy barb, serum from purple-headed barb, black-spot barb and spanner barb do not display com-plement activity (Rijkers & Van Muiswinkel, 1977). In the plaque assay of carp, serum of goldfish is ineffective (Rijkers et al., 1980a). It is therefore unlikely that functional interchangability of complement can be used as a taxonomical criterium.
Fish complement is present not only in serum. In rainbow trout a heatlabile
anti-Vibrio activity was observed in mucus, presumably complement + antibody (Harrell, Etlinger & Hodgins, 1976).
Only few data exist on isolated components of fish complement. Ross & Jensen (1973b) have isolated and purified the first component (C1n) of the complement system of nurse shark. C1n was incompatible with rabbit immunoglobulin but formed an inter-mediate complex with sheep erythrocytes sensitized with nurse shark antibody. These intermediates could be specifically lysed with guinea pig serum devoid of C1. On the other hand, whole guinea pig complement did not react with SRBC sensitized with shark antibody. It was concluded that the first component of shark complement can initiate the cascade reaction of the mammalian complement system provided that it is allowed to react with an immune complex containing a compatible antibody (Ross & Jensen, 1973a). In addition Gigli & Austen (1971) have demonstrated the presence of C9 in nurse shark serum which reacted with the cellular intermediate EAC 14235678 prepared with guinea pig serum.
Cobra venom factor (CVF) depletes the terminal complement components by activat-ing the complement sequence at C3. The complement activatactivat-ing effect of CVF requires the cooperation of the alternative pathway system. This alternative complement activi-ty was present in hagfish serum and in hemolymph of horseshoe crab (Limulus polyphemus)
and in sipunculid worm (Golfingea sp.) but absent in lamprey, nurse shark, paddlefish and carp sera (Day, Gewürz, Johannsen, Finstad & Good, 1970). Classical complement activity was not observed in hagfish, lamprey, horseshoe, crab and sipunculid worm. These findings suggest that the terminal components of the complement system, which can be activated without antibody, appeared early during evolution. A complement system, which requires antibody-mediated activation is only found in the vertebrates were immunoglobulin has been demonstrated.
b) Lysozyme
Lysozyme, an enzym with bacteriolytic properties, is present in serum, mucus and phagocytic cells of many fish species (Luk'yanenko, 196S; Fletcher & Grant, 1968; Fletcher & White, 1973b; Ourth, 1980). The molecular weight (15 x 10 ) is similar to mammalian lysozyme. The different electroporetic mobility reflects differences in amino acid composition (Fletcher & White, 1973b).
