Speciall events contributed to putting the pieces of the puzzle in right order. The potassiumm channel conference in Ulm, where 1 met prof. Clifford Slayman (Yale), whoo selflessly provided advice, encouragement and insightful discussions. Another off Richard Adrian's students, Prof. CL Huang (Cambrigde), was helpful in discussingg skeletal muscle physiology. The physiological conference in Prague, wheree I met prof. ADC Macknight (University of Otanogo), who helped in sorting outt the issues concerning Chloride. The courses mathematical modelling by Prof. H Westerhoff,, the ERC course by Prof. DL Ypey (LUMC) and the QuB course by Profs Auerbach,, Qin and Sachs (State University of New York), where I also met drs. Grossmann (University of Illinois) and Milescu. These five individuals challenged me intoo sorting my thoughts about data analysis and to analyse what is observed, not whatt might be observed with a perfect instrument or data set. In this particular regardd Lorin Milescu (NIH) was very forthcoming in sharing his thoughts and knowledge. .
Dr.. Jan Siegenbeek van Heukelom hired me and delivered the necessary means to completee the thesis. I enjoyed the freedom Prof. Wytse Wadman provide. Dr. Harald vann Mil was very important at every twist and turn during the process.

I am grateful Prof. CMA Pennartzz was forthcoming in judging the manuscript. Prof. Cornelissen and Dr. JP de Jongg made special contributions by steering the process in the right direction. In the labb Drs. J Groot, R Lingeman, W Ghijsen, W Raamsdonck, T Werkman, S Nair and W Hesenn took part in interesting discussions, ranging from curry to microscopy. 1 humblyy appreciate the opportunity offered by the editors of The Journal of Physiologyy for inviting me to act as a recurring referee.
II enjoyed being distracted by Roderik and René and other basketball players, and by playingg squash with fellow bursalen Marco Roos, Martijn Egas and José Vos.
