1. Multicomponent reactions (MCRs) are a useful tool in combinatorial chemistry and medicinal chemistry.
2. MCRs are an advanced class of organic reactions which has facilitated rapid access to diverse libraries of biologically important molecules because of its ease of synthetic operation.
3. The sequencing of MCRs and subsequent modifications is one of the most successful strategies leading to high structural diversity and molecular complexity.
4. Ugi reactions employing ammonia as the amine component offered a simple, one-step assembly of Ugi adducts suitable for elaboration into a variety of complex structural frameworks.
5. 1,3,4‐Oxadiazoles are a kind of aromatic heterocycles valued for their low-lipophilicity in drug development and exhibit a variety of biological effects.
6. Transition-metal-catalyzed annulation reactions have been extensively investigated in post-Ugi transformations, especially in the context of constructing multiply functionalized nitrogen (N)-containing (poly)heterocycles.
7. Targeted protein degradation using Proteolysis Targeting Chimeras (PROTACs) has emerged as a novel therapeutic modality in drug discovery and attracted the favor of academic institutions, large pharmaceutical enterprises, and biotechnology companies.
