242 Firstly, I would like to thank my promotor Thorben Cordes. Thank you for all your help, for all the opportunities you gave me, for involving me in various projects and especially for the freedom you gave me to explore my own interests. It was a great learning experience.

Giorgos Gkouridis, thank you for your help and all the years we worked together. I cherish

the scientific and non-scientific discussions we had. I also would like to thank my second promotor Erik van der Giessen, especially for the suggestions on the theoretical work of this thesis.

I would like to thank my assessment committee, Philip Tinnefeld, Enrica Bordignon and Wouter Roos. Thank you for critically reading this thesis.

The work of this thesis was done in collaboration with many researchers from other groups and universities. I want to express my gratitude to them and I am proud that our work resulted in many publications. Gea Schuurman-Wolters, Ruslan Vietrov, Martyna

Pietrzyk, Marco van den Noort and Bert Poolman and all other members of the

Enzymology group: I appreciated the fruitful collaborations we had. Especially

Bert Poolman, thank you for always taking the time for me and for involving me in many

projects. I also would like to thank Stephanie Begg and Christopher McDevitt of the University of Melbourne and Bianca Hetzert, Kristin Kiosze-Becker, Holger Heinemann,

Elina Nürenberg-Goloub and Robert Tampé of the Goethe University Frankfurt.

I also would like to thank all of my direct colleagues for their help in the lab and for the discussions we had, especially my close collaborators Kostas, Florence and Yusran. Thank you for all your help! I would also like to thank the other members of the Molecular Microscopy group whom I worked closely with for many years: Nikos, Atieh, Evelyn,

Jochem, Jasper, Matthijs, Martin and Yichen. I also want to thank the members of the

Molecular Biophysics group, Sourav, Melissa, Guus, Pedro, Ceri, Ignacio, Yuzhen and

Wouter and the people of the Single-Molecule Biophysics group, Jelle, Karl, Andrew, Joris, Sarah, Sameneh, Alex and Antoine.

Victor, your support and advice on the optics was essential; thank you for sharing your

experience with me. Margriet, thank you for your help, not only on organisational related issues. Michiel, thank you for your support on computer related issues.

I also would like to recognize the facilities provided by the Zernike Institute for Advanced Materials, and want to thank especially Caspar van der Wal and Jan Peter

Birkner.

Last but not least, I want to express my gratitude to Monique. Thank you for your endless support!

By using single-molecule FRET we show that the two ATP sites of ABCE1 are always in a dynamic equilibrium between three distinct conformational states: open, intermediate and

When only the energy of

To circumvent this limitation, and to directly relate the SBP conformational changes with the binding of substrate, we developed a single-molecule assay in Chapter 5 to

Dus de NBDs van ABCE1 switchen niet tussen een open en dichte conformatie maar kunnen vele verschillende conformaties aannemen door de asynchrone bewegingen in beide

State 1: free ABCE1 sites are in an equilibrium between three states (open, intermediate and closed), but predominantly found in the open conformation.. State

The spontaneous conformational changes in SBPs, ABCE1 and other proteins influence the conformational equilibrium when a ligand is bound as well as the affinity between the protein
