Molecular analysis of circadian photosensitivity and diapause in the jewel wasp Nasonia vitripennis

Marcela Buřičová

1. The pattern of cryptochrome2 (cry2) expression may be more important than its level of expression, as circadian rhythms are maintained even after reduced levels of cry2 mRNA (this thesis).

2. As Nasonia CRY2 is not light sensitive (this thesis), further investigation of proposed candidates for photoreceptors involved in light-entrainment of the clock in Nasonia is urgently needed.

3. The circadian clock regulates the switch point for diapause induction, but this is only the beginning of a complex trait with many regulatory

pathways.

4. Like life, research can always bring unexpected results to deal with. 5. Not only positive results are important results.

6. Asking good questions is just as important as giving good answers. 7. "The methods of science are not fool-proof, but they are indefinitely

perfectible. Just as important - there is a tradition of criticism that enforces improvement whenever and wherever flaws are discovered".

We did not question the involvement of the circadian clock in seasonal timing measurement, which was shown before in Nasonia (Saunders 1970, 1974), instead we made an attempt to

Strong circadian light sensitivity and a broad phase angle of entrainment can explain photoperiodic response in different T-cycles by applying the external

Variation in circadian free-running period in Nasonia vitripennis is necessary for adaptation of seasonal diapause induction to different latitudes (Chapter 2). The

This different timing of activity reflects the speed of the clock in constant darkness (DD): southern lines show shorter free running rhythms (τ) close to 24h (faster

For the clock genes period (per), chryptochrome-2 (cry-2), clock (clk) and cycle (cyc), circadian expression depending on photoperiod and latitude of origin was analysed
