1. Targeting TNF superfamily ligands or receptors with either agonistic or antagonistic compounds may provide novel approaches for therapy (this thesis). 2. The combination of Computational Protein Design and focused mutagenesis with

screening is a powerful approach to modify protein-protein interactions (this thesis).

3. A single amino acid substitution can cause a great impact on protein-protein interactions (this thesis).

4. RANKL mutants with reduced binding to OPG show potential anti-fibrosis activity, which offers a promising strategy to explore new therapeutics against fibrosis (this thesis).
