Een fantastische, spannende en leerzame tijd in Groningen gaat nou op voor mij. The work on this thesis in the Single Molecule Biophysics department was both a challenge and rewarding experience for me. Luckily, I have been surrounded by people who supported me and made my PhD research projects possible.

Dear Antoine, thank you for your support and patient guidance during my PhD. It was a pleasure working and developing research projects with you. You have created an inspiring and encouraging atmosphere in the SMB group in Groningen, which I always enjoyed being part of. I appreciated a lot that you always kept your door open to discuss any problems, ideas, and results.

I am very grateful for the evaluation of my thesis by the members of the assessment commitee, Prof. Diez, Prof. Maglia, and Prof. Claessens.

I would also like to thank our collaboration partners: Prof. Dixon and Dr. Zhi-Qiang Xu from Wollongong who helped tremendously with the E. coli replication project; Prof. Charles Richardson and Dr. Arek Kulczyk from Boston for their advice on the T7 replication proteins; Prof. Sohn and Dr. Seamus Morrone from Baltimore for the collaboration on the IFI project; and Prof. Chiechi and Prof. Verpoorte from Groningen, with whom I had the pleasure to work on the nanowire-based chip within a highly interdisciplinary project. Many thanks for the numerous ideas and suggestions in the course of our projects!

My dear paranymphs, Evelyn and Jan-Peter, I’m very happy to have you next to me during my defence. Evelyn, you have helped me in any possible optics question, and always had time to listen to my bigger and smaller problems. And, of course, thanks for your help with the cover! Jan-Peter, thank you for all the advice in the beginning of my PhD; I profited a lot from your molecular biology tips! You both accompanied me for large portions of my PhD, both in the lab and outside. I’m very grateful to have met you in Groningen.

I would also like to thank Thorben and Wouter for providing a supporting atmosphere to the SMB lab remainders during the last year.

My dear replication sub-group members, Hylkje, Audrey, Karl, Lisanne, and Enrico, thank you for the many ideas and inputs during our weekly meetings which helped me a lot struc-turing and developing my research projects! Good luck, Lisanne and Enrico, with finishing your theses down-under. Machteld, it was a pleasure supervising you during your master. Good luck with your PhD research!

My dear (last) office mates, Florence, Atieh, Kostas, and Alex, I was very lucky to share the office room with you. It was always fun talking about projects and many other things outside Zernike.

Many thanks to our biochemistry experts, Guus and Giorgos, for highly valuable advice on any problem I encountered in the wet lab.

Dear Victor, I’m very grateful for your help with all my microscope issues and during the last year with the microscope moving organization. Dear Michiel, many thanks for the frequent support with Fiji macros!

I would like to thank past and current members, as well as close collaborators of the van Oijen lab and the Cordes lab for creating a constructive, motivating spirit during work, lunches and conferences: Andrew, Harshad, Victor, Da, Flynn, Jochem, Jasper, Marijn, Monique, Yichen, Muhammed, LJ, Joris, Ruslan, Jason, Jelle, Samaneh, Marysia, Ilja, Nadine, and Iulia, it was a pleasure working with you.

I would also like to thank Jeannette and Margriet, who helped me so much in organizing the start of my PhD and the final phase.

Luckily, Nijenborgh 4 provided very helpful neighbouring groups that helped me quite of-ten with expertise and lab equipment. Dear Nina, Ana, and Gosia, thanks for helping out with various expression systems, for discussions on the corridor and parties. Dear Gea, many thanks for correcting my dutch translation! It was a pleasure joining the settlers evenings with you and Karin. I would also like to thank Pavlo for the aquisition of the AFM data, Ria for help with immunoblot assays, Wiktor for discussions and help with unnatural amino acid labeling, Alicja for mass-spec support, and Sonja for many helpful discussions.
