Acknowledgements

Bert.It took you one minute to agree with me when I asked to start a PhD although you had to move around with financials and all other sorts of stuff. Even today, I still don’t know why this decision was made so quickly, there was no position available and you had a need for supporting staff. I like to think this is perhaps just how you are. A person that when he sees a tiny bit of potential he is willing to provide the means for that person to flourish. Obviously, I can’t thank you enough for everything you’ve done for me these last seven years. Scientifically, but even more the personal discussions we had in which I was able to ask you almost anything. With that, to me, you are an incredible warm person with a great heart that cherishes personal relations and invests in them like friends would do.

Dirk-Jan. I find it fantastic to be in your presence, you’re energetic, optimistic and open. You probably talk even more than I do and you don’t mince words. Because of that I always had the urge to tell you my latest results, even though Bert hadn’t seen them yet. This brought me many new ideas and insights that ultimately led to where I am now. Frans. Now you’re in my acknowledgements. You were there from the start and are sort of still here when I finish. Started as my supervisor we’ve become close friends and I started a PhD with the foundation you’ve laid out for me. With that, you’ve been far more for me than a supervisor and close friend. I can’t thank you enough for everything you’ve given to me.

Hendrik. Colleague, friend and paranymph. We started our PhD nearly at the exact same moment and are probably finishing the same year as well. We’ve became good friends and you’ve supported me along the way. mostly with some personal advice, but also with some essential scientific contributions to my research towards the end of my PhD. Thank you for always being there for me, at any time and any place.

Ria. Mijn kantoor maatje. Zittend achter mij, wist ik altijd wat er gaande was in het lab. Je had een fantistisch gevoel voor mensen en wat hen motiveert. Je schroomde je niet om taboes te doorbreken en stelde soms de vraag die niemand durfde te stellen. Deze houding en mentaliteit waardeerde ik enorm en heeft mij vele inzichten gebracht die ik tot de dag van vandaag meeneem in mijn denken en doen.

- 197 - Gea. I admire your expertise, everlasting energy and holy molly, you are able to take on a dozen of tasks without forgetting even one. I loved trying to make you frustrated about little things and see you realizing I did that on purpose. The tomato head you then get in combination with shaking you head while smiling. Unforgettable for me! I hope we will see each other somehow later on. Besides, you will probably never make a bet with me again, but I have to ask the question once more. When are you planning on finishing your thesis?

Bauke & Tjeerd. You both started a little bit earlier than I did, but you guys, together with Hendrik are the group of dutchies that I commenced my PhD with. I enjoyed the many useful and sometimes not so useful discussion we had. Especially the l ones we had during the first two years of our PhDs at the Friday afternoon drinks. I mostly remember those as I really needed them due to many experimental failures I had at the start. Thank god, you guys were there for me back then!

The yeast group: Gemma, Stephanie, Ryan, Lukasz, Alex, Evi and Jacopo. Some of you were there from the beginning while others finished or have yet to finish. In any case, all of you played a different part. Gemma and Stephanie from the beginning as guidance and teaching me all the ins and outs. Ryan and Lukasz, you guys were sort of my sparing partners in all of this. Challenging each other with the main questions of our research and providing scientific support whenever I needed it. Alex, Evi and Jacopo, you actually witness the final steps of my PhD, but also you played a significant role towards the end by providing a great ambiance within the small yeast group and giving me that last push to finish.

Finally, the students; Jarnick, Ruben & Mirco. The chapter and publication on the extracellular loops would not be there without the diligent efforts of the three of you. I very much enjoyed not only supervising you all during your master projects, but also the personal connections we shared at that time.

Branch & Tan-Yun. My collaborators from Harvard medical school. Your expertise in lipid mass spectrometry is astonishing and it brought chapter 4 to a whole other level. Together, we made great observation that led to a more complete model of the yeast plasma membrane. I especially appreciated the scientific discussion mixed with small jokes. This made working together very pleasant and gave me the feeling of being able to communicate about our work as it should be: transparent, critical with an eye for detail.

Laetitia. Of course, I can never forget our first encounter. You were assigned as a master student to Ryan, almost not speaking a word of English and my French was similar to my understanding of Mandarin. Yet, you were able to catch my eye and we felt a connection instantly. During the first weeks of that summer we became close friends, but that friendship quickly became more. At that time, you moved back to France to finish your studies and we could only see each other every two weeks somewhere halfway in Belgium. Despite the distance, this was a period for both of us with many highs and basically no lows. Now, 2.5 years later. We are living together for almost 1.5 year and are searching for a life somewhere across the north Atlantic Ocean. You bring me optimism, joy, laughter and love at all times, but more so when I unconsciously need it. I often realize later that it is actually you who give me that final push to succeed in whatever I am doing. I can’t imagine starting a new adventure abroad without you. Perhaps a cliché, but words do not exist to express what you mean to me. C’est dans la poche!

Familie. Wij zijn allen opgegroeid in het midden van Nederland, in een klein dorpje ‘Achterveld’, nabij Amersfoort. Hierdoor blijft Groningen altijd een eindje reizen in jullie ogen en zien wij elkaar minder vaak dan wij zouden willen. Deze afstand en ook de spreekwoordelijke afstand tussen het type werk dat ieder van ons doet maakt het soms lastig om diep begrip van elkaars leven te hebben. Ondanks dat, heeft ieder van jullie mij te allen tijde gesteund in alles wat ik doe. Soms twijfelend met kritische vragen, maar uiteindelijk is er altijd begrip voor de keuzes die ik maak. Deze steun en toeverlaat is voor mij van onschatbare waarde en zal van belang blijven bij elke stap die ik mijn leven ga zetten.

I would like to thank all members of the Enzymology and Cryo-EM group for the discussions and practical assistance I enjoyed being a member of this amazing group of intelligent and kind people.
