ACKNOWLEDGEMENTS

The completion of this thesis would not be possible without help from many people, and now I would like to take the opportunity to thank everyone who supported me along the way.

First of all, I would like to thank my mentor and co-promotor. Kris, I am very grateful for your guidance and understanding during all these years - it was a fantastic lesson and I have learned a lot from you. You provided me with a perfect balance between supervision and freedom - the combination which, I believe, was the key to the success of this project. Despite my tendency to focus on negative aspects of my work, you were always able to force me to notice the bright sides of it as well. Thank you for your faith in my scientific skills - I would not have developed them without your continuous support and patience. Even though this 'chapter' is now almost over, I hope we will have a chance to work together in the future again.

Next, I would like express my gratitude to the promotor of this thesis, Paul-Peter. Thank you for giving me a chance to work in your department during my masters internship, which has subsequently developed into a PhD project. Without your initiative and management, this work would not have been possible. Although your travel obligations often limited our chances to discuss my work in person, it has been reassuring to know that, when needed, I could always count on your timely support from afar.

I would like to thank my dear paranimfen. Linda, working with you in the lab was a great experience and I learned a lot from it. Your organizational skills and your drive to work with the highest possible efficiency were always very motivating. I also really enjoyed our long discussions after work over a glass of Duvel – they were inspiring in many different ways. Inês, your everlasting enthusiasm and positive energy made late evenings in the lab a lot nicer. Thank you for all the fun time we spend together outside of the lab, and for your help with preparations for my promotion. And finally, thank you for your smile, which makes the whole K0 corridor a much better place!

This work would not be possible without help from my colleagues in the group. Joana, Daphne, Sarah, Marjolein, Wendy, Debby, Iris, Inge, Samuel, Bea and Chiara, thank you for your help in the lab and critical approach during work discussions. I learned a lot from all of you! It was both pleasure and privilege to work together! Dion, even though we worked as a team only for a couple of months, it was an extremely efficient period and I truly regret that we didn't have a chance to work together a bit longer.

I would like to express my gratitude to Daan Gerlag, Man-Wai and Tamara, as well as all the KIR clinicians for providing me with patient material - without your continuous support this work would not be possible. Lisa, your expertise in gene expression analyses was very helpful - thanks for sharing your knowledge and data with me.

I would like to thank Eric, Jörg, René Lutter and Ronald van Ree for helpful suggestions and discussions. Dominique, thank you for all your questions and comments after my presentations - they always allowed me to look at my work from a different perspective and see possibilities that I overlooked before.

I would also like to acknowledge our collaborators from the University Hospital Zürich: Caroline Ospelt for fruitful collaboration and inspiring discussions about studying epigenetics in RA, and Prof. Steffen Gay for joining my thesis committee and for reviewing this thesis.

This work would not be possible without long-term collaboration with Italfarmaco S.p.A. I would like to thank Dr. Paolo Mascagni for providing us with ITF2357 and supporting our research.

I would like to thank all the roommates I had during my stay at the AMC: Cristina, Fernanda, Lara, Simone, Daan, Margot, Henrike, Kirsten, Marjolein. Thank you for creating a nice atmosphere in the office and for all the funny, relaxing conversations.

Olga, I know that you were sometimes upset by my jokes, but I am happy that you eventually got used to them - being your neighbour in the office for the last couple of years was a lot of fun. Dennis, thanks for being a great companion during your stay at the AMC. Your unique sense of humour made our lunch and coke breaks really fun. Also, thanks for teaching me how to play pool - I enjoyed our pool sessions after work tremendously!

Gabriela, Tineke, Felix, Sven, Flavia, Troy, thank you for all the nice moments outside of the lab, especially in the "Oude Gasthuis" - I truly enjoyed them!

Paweł, dzięki ze zdecydowałeś się dołączyć do naszej grupy – Twoja obecność uczyniła ostatnie miesiące znacznie mniej stresującymi. Powodzenia z doktoratem!

Thank you… Bedankt… Dziękuję! Alek
