(3) All great things have to come to an end, even this thesis :) Now it is time to thank all the people without whom this journey would not have been possible or nearly as fun. First and foremost, Marthe – it is an honour to be promoted by you (now Prof. Walvoort!) and I could not wish for a better supervisor. While the research was often difficult, I enjoyed navigating it with you. Thank you for your unwavering support and understanding. I am proud to be an alumnus! Adri, thank you for always putting in extra effort to understand biological side of things in my research. I appreciate all the advice and feedback over the years from you. Martin, thank you for all the discussions and thoughtful advice, no matter the subject. Big thanks to the members of the reading committee for your efforts and helpful feedback! To Anna, Gianluca, Alexander, Daniel and Zeynep - thank you for the experimental efforts and for providing a great mentoring experience. It is inspiring to see other people grow and I am sure each of you has a bright future ahead! To many collaborators without whom I wouldn’t know half of the things I know now. First off, to Prof. Nathaniel Martin and Tom Wood who bravely joined us on the EarP project. Thank you for always being enthusiastic and incredibly hard-working! To Johan Kemmink: one can only aspire to become an expert of such level, thank you for being on our team! Ioli, our ITC-fairy, thank you for joining the project, it was superfun to work together! To our German collaborators: Prof. Lassak, Franziska and Ralph, thank you for reaching out, it was great to connect and accomplish results together. For the semiprocessivity project: big thanks to Carlos and Prof. Siewert-Jan Marrink for adding your expertise to our project. It benefited immensely from it and we learnt so much! It was a complex system to navigate and I am grateful for your efforts to understand it. To Robbert Cool for the help with challenging SPR analyses. To Johan Hekelaar – thank you for all the help with the proteomics analysis over the years and willingness to answer my never-ending questions :) To Hjalmar Permentier, Walid Maho and Marcel de Vries from the Interfaculty Mass Spectrometry Center – thank you for all the samples analyzed (the count . 241.

(4) was in the hundreds, I am sure) and your expertise in the data interpretation and analysis. To everyone involved in the oxidation project, directly or indirectly. Niels, I appreciate your involvement very much! You are one of the most methodical and dedicated people I have met and I am grateful to have worked beside you. To talented researchers involved in the azidoglucose-labeling project: Zeynep for your incredible efforts in the chemical synthesis! Feliciana, Francesca and Prof. Anna Maria Papini thank you for your expertise in the antibody-assays and exciting discussions! To Aike Jeucken, Prof. Dirk-Jan Scheffers and Prof. Dirk Slotboom for the help with elucidating the multimeric structures of the N-glycosyltransferases. Thank you for your time and efforts, this was a very fun spin-off project and I am excited to see what else is there to the story! To Roderick, Florian and Prof. Giovanni Maglia for our collaboration on the nanopore project. It is an exciting new horizon in science and I am happy to have participated in the research about it! To all the support staff: Paulien for your resourcefulness and a wealth of technical knowledge! To Hilda and Janka for your organizational efforts and coordination! To Monique Smith, Johan Kemmink, Peter van der Meulen, Johan Hekelaar, Franz Ho and Renze Sneep for keeping all the machines running smoothly and your help with analysis! To my friends inside and outside of academia: Jana, thank you for being my paranymph, it has been a lot of fun to do our PhDs together and I know you will go on to achieve great things! To Isabel – thanks for all the laughs and GTs, girl! I know you will find a great place to apply your talents! To Anastasiia, Maria and Olya: thank you for all the support and understanding over the years. I am excited to see what you will accomplish in the future and I hope to be there to celebrate! To all the past and current members of the Walvoort group and Chemical Biology group. No matter the composition, Chemical Biology group is always an exciting and stimulating environment to be, and it will stay like that in my memory. .

(5) To Tom: thank you for making my life so much brighter and sweeter. You are truly a superstar and I can’t wait to spend the rest of time with you. KH:]. . 243.
