Compartmentalized cAMP Signaling in COPD

Focus on Phosphodiesterases and A-Kinase Anchoring Proteins

By Haoxiao Zuo

1. Measuring FRET signals in PCLS is a novel approach to monitor cAMP dynamics on the level of the tissue (this thesis).

2. Although both PDE3 and PDE4 are responsible for cAMP hydrolysis, the different functional outcome (ciliary beating versus airway smooth muscle relaxation) by inhibition of PDE3 and PDE4 can partly be explained by the distinct distribution of PDE3 and PDE4 in different cell types (this thesis). 3. The expression (mRNA and protein) and activity of both PDE3 and PDE4 are upregulated by cigarette smoke exposure, which likely underpins alterations of intracellular cAMP levels. Therefore, dual inhibition of PDE3 and PDE4 shows a promising benefit in cigarette smoke-induced lung disorders (this thesis).

4. AKAPs, especially Ezrin, AKAP95 and Yotiao, are important to maintain TGF-β1/ cigarette smoke-induced epithelial-to-mesenchymal transition. Therefore, Ezrin, AKAP95 and Yotiao complexes may represent a novel target to reduce TGF-β1/cigarette smoke-induced epithelial-to-mesenchymal transition (this thesis).

5. Take heart and be strong; have no fear and do not be troubled; for the Lord your God is with you wherever you go. --- Joshua 1:9.

你当刚强壮胆。不要惧怕,也不要惊惶。因为你无论往哪里去,耶和华你 的神必与你同在。---约书亚记 1 章 9 节。

6. Doing research and raising a kid share some common points: being creative, being thoughtful, being patient, and being happy.

7. Success is going from failure to failure without losing enthusiasm. 8. Life is like a box of chocolates, you never know what you are going to get. --- Forrest Gump

Targeting domains (TD) are protein components of various multiprotein complexes which when fused to the sensor, direct the sensor to the subcellular site where the targeting domain

Similarly, it was reported earlier that the expression of ezrin was subject to alterations in both airway smooth muscle exposed to cigarette smoke and lung

The co-silencing of Ezrin, AKAP95 and Yotiao inhibited TGF- β1-induced cell migration in BEAS-2B cells and primary human airway epithelial cells.. In addition, co-silencing

The objective of this thesis was to investigate the role of cAMP compartmentalization in COPD, mainly focusing on phosphodiesterases (PDEs) and A-kinase anchoring proteins
