1. Proposition 1. - Energy is what keeps systems out of equilibrium, therefore its sup-ply should be the first and most important design aspect of a syntetic cell. (Chapter 1)

2. Proposition 2. - Understanding a system globally does not imply understanding of the details, and vice versa. Both are of importance when building a system from scratch. (Chapter 2)

3. Proposition 3. - Cyclic-di-AMP acts as an emergency brake in the ABC-transporter OpuA. (Chapter 3)

4. Proposition 4. - Theory and in vitro / in silico approaches become equivalent to the in vivo situation when we have a full understanding of the cell. Until then a seemingly simple modification, e.g. turning a homodimer into a heterodimer, remains a significant challenge. (Chapter 4)

5. Proposition 5. - Time investments in the present, often save time in the future. (Chapter 5)

6. Proposition 6. - Failures bring the knowledge, success is needed to keep one moti-vated.

7. Proposition 7. - Problem solving is interesting and exciting, having problems is not.

8. Proposition 8. - Collaboration multiplies either frustration or success, depending on the communication and amount of energy put in by all parties.

1. Stelling 1. - Energie houdt systemen uit evenwicht, daarom zou energievoorzie-ning het eerste en meest belangrijke ontwerp onderdeel van een synthetische cel moeten zijn. (Hoofdstuk 1)

2. Stelling 2. - Een systeem globaal begrijpen betekent niet dat we de details begrij-pen en vice versa. Beide zijn van belang wanneer een nieuw systeem ontworbegrij-pen wordt. (Hoofdstuk 2)

3. Stelling 3. - Cyclisch-di-AMP fungeert als een noodrem op de ABC-transporter OpuA. (Hoofdstuk 3)

4. Stelling 4. - Theorie en in vitro / in silico benaderingen worden equivalent aan de

in vivo situatie wanneer we de cel volledig begrijpen. Tot dat moment zal een op

het oog eenvoudige aanpassing, zoals het veranderen van een homodimeer in een heterodimeer, een aanzienlijke uitdaging blijven. (Hoofdstuk 4)

5. Stelling 5. - Het investeren van tijd in het nu leidt vaak to besparing van tijd in de toekomst. (Hoofdstuk 5)

6. Stelling 6. - Mislukkingen brengen kennis, succes is nodig om gemotiveerd te blij-ven.
