Inearly2002,whenIwasfinishingmyMScinBiotechnologyatWageningenUniversity,I

wanted to further broaden my knowledge on molecular biology, especially in the field of

functional genomics. An interesting PhD position was available from the fungal IOP

program.IappliedforthisandafewmonthslaterIstartedmyPhDprojectattheInstitute

ofBiologyofLeidenUniversity(IBL),inthefungalgeneticsgroupofProf.vandenHondel.

Now,afewyearslater,withalotmoreknowledgeandalotmoreexperience,the

project is finished. This thesis is the final tangible result. The project itself, however, was

enjoyable, of course with its difficult moments but I look back on a very nice period in

whichIdevelopedmyselfalot,bothprofessionallyandpersonally.

It would not have been possible to finish the project without many contributions, both

scientifically and personally. First of all, I want to thank the fungal IOP program for

financiallysupportingmyPhDproject.Thiswasessentialforthesuccessfulcompletionof

theproject.

In the first place I would like to thank my colleagues at the fungal genetics group at the

this fresh colleague, both with her experiments and with her personal life. Your kind

assistance made the project going more smoothly. Davy, without you the lab would lose

AmyR. Adjacent to the gene for the AmyR transcription factor are genes encoding

functions of the other predicted family GH13, GH15 and GH31 enzymes, including.. intracellular enzymes and cell wall associated proteins, in alternative

sucrose act as an inducer for the inulinolytic genes and we therefore propose that the. sucrose which is formed from inulin or raffinose induces expression

activate the expression of the structural genes in the presence of inducing molecules. In. Chapter 7 we describe the identification and characterization of
