Targeting hepatic stellate cells to prevent or reverse liver
fibrosis
1. Hepatic stellate cells are the main precursors of myofibroblasts in the pathogenesis of liver fibrosis. Targeting hepatic stellate cells is therefore an efficient way to prevent or reverse liver fibrosis. (This thesis)
2. Induction of hepatic stellate cell senescence can attenuate liver fibrosis. However, senescence inducers should be combined with senolytics to avoid the side effects of senescence. (This thesis)
3. The coumarin derivative esculetin can induce senescence of hepatic stellate cells and decrease fibrogenesis. The mechanism of esculetin is dependent on PI3K-Akt-GSK3β signaling pathway. (This thesis)
4. Endogenous production of H2S is essential for the activation and proliferation of hepatic stellate
cells. Cystathionine γ-lyase (CTH) is the major enzyme responsible for the synthesis of H2S in
activated hepatic stellate cells. (This thesis)
5. Inhibition of cystathionine γ-lyase (CTH) induces senescence of hepatic stellate cells and reduces proliferation and collagen synthesis. (This thesis)
6. The broad-spectrum anti-fibrotic drug Pirfenidone reduces cell proliferation and collagen production of fibroblasts via the TGF-β/mTOR axis.
7. During the activation of hepatic stellate cells, the expression of the arginine metabolizing enzyme Arginase-1 (ARG-1) is increased whereas the expression of the arginine metabolizing enzyme Inducible Nitric Oxide Synthase is decreased. Targeting ARG1 in activated hepatic stellate cells reduces fibrogenesis because it will prevent the synthesis of proline precursors. (This thesis)
